摘  要:目的:探讨植物生长调节剂、基本培养基、蔗糖质量浓度以及相关添加物对铁皮石斛茎段原球茎的诱导、增殖、分化以及根部生长等方面的影响。方法:通过正交试验方式,要求没有添加激素下,对铁皮石斛茎段原球茎进行消毒处理、试验培养基的配备、对原球茎进行诱导、对原球茎进行增殖和分化处理、生根的培养等,分析不同植物生长调节剂、基本培养基配比对于茎段原球茎的诱导影响,不同基本培养基、马铃薯泥以及蔗糖浓度增殖分化原球茎效果,以及香蕉泥、萘乙酸(NAA)和马铃薯泥对铁皮石斛茎段原球茎生根效果。结果:1)最适宜诱导铁皮石斛茎段原球茎的培养基组合是NAA 0.1 mg/L+1/2MS(Murashige and Skoog)培养基+6-苄氨基腺嘌呤(6-BA)4 mg/L。2)铁皮石斛茎段原球茎增殖最佳的培养基是:MS+蔗糖30 g/L;分化的最佳培养基是:马铃薯泥10%+蔗糖10 g/L+1/2 MS。3)铁皮石斛茎段原球茎的生根最佳培养基组合是马铃薯泥10%+1/2 MS+NAA 1 mg/L+香蕉泥10%。结论:在没有添加激素的前提下,基于正交试验方式发现变化蔗糖含量和基本培养基能够对铁皮石斛茎段原球茎增殖和分化效果进行调节,并找出了最理想的组合模式。Objective:To investigate the effects of plant growth regulators,basic medium,sucrose concentration,and related additives on the induction,proliferation,differentiation,and root growth of protocorm from stem segments of Dendrobium candidum.Methods:According to the requirements of the orthogonal experiment,the protocorm from stem segments of D.candidum was sterilized without adding hormones,and the test medium was prepared.The protocorm was induced,proliferated,and differentiated,and the root of the protocorm was cultivated.The effects of different plant growth regulators and basic medium ratios on the induction of protocorm from stem segments were analyzed.The effects of different basic media,potato mash,and sucrose concentration on the proliferation and differentiation of protocorm were analyzed,as well as the rooting effects of banana mash,naphthyl acetic acid(NAA),and potato mash on the protocorm from stem segments of D.candidum.Results:1)The most suitable medium combination for inducing the protocorm from stem segments of D.candidum was 0.10 mg/L NAA+1/2 Murashige&Skoog(MS)+4 mg/L 6-benzylaminopurine(6-BA).2)The optimal medium for proliferation of the protocorm from stem segments of D.candidum was MS+30 g/L sucrose,and the optimal medium for differentiation was 10% potato mash+10 g/L sucrose+1/2 MS.3)The optimal medium combination for rooting the the protocorm from stem segments of D.candidum was 10% potato mash+1/2 MS+1 mg/L NAA+10% banana mash.Conclusion:Without adding hormones,the results of the orthogonal experiment show that the changes in the sucrose content and the basic medium can regulate the proliferation and differentiation of the protocorm from stem segments of D.candidum,and the most ideal combination mode has been found in this study.
