摘 要:目的:观察藏药七味铁屑(QWTX)方对非酒精性脂肪肝病(NAFLD)SD大鼠脂质代谢的影响,为临床用药提供理论依据。方法:选取90只SD大鼠随机分为正常组、模型组、西药组,QWTX高剂量组、QWTX中剂量组、QWTX低剂量组,每组15只。正常组予以普通饲料喂养,其余各组予以高脂饲料喂养。喂养3周后,各观察组给予相应药物进行干预,并每周逐一称重调整药物用量,持续6周。第9周末,检测血脂、游离脂肪酸(FFA)、一氧化氮(NO)含量;肝组织匀浆检测超氧化物歧化酶(SOD)、丙二醛(MDA)含量,HE染色观察肝细胞脂肪变程度。结果:1)病理显示,模型组大鼠肝细胞内多呈大泡样混合型脂滴,伴散在炎症细胞浸润;QWTX各剂量组可降低肝组织脂肪含量,抑制脂质沉积;2)血脂四项QWTX各剂量组与模型组比较,差异有统计学意义(P<0.05);3)与模型组比较,QWTX各剂量组降低MDA水平、FFA含量,升高SOD水平、NO含量(P<0.05)。结论:藏药QWTX可改善NAFLD脂代谢,作用机制可能与抗脂质过氧化反应有关。Objective:To observe the effect of Qiwei Tiexie(QWTX)Prescription on lipid metabolism in experimental SD rats with non-alcoholic fatty liver disease(NAFLD),and to provide theoretical basis for clinical use of QWTX.Methods:Ninety SD rats were randomly divided into normal group,model group,positive drug group and QWTX high-,medium-and low-dose groups,15 in each group.The normal group was fed with normal diet,while the rest groups were fed with high-fat diet.After 3 weeks of feeding,each treatment group was given corresponding drugs for intervention,and the rats were weighed one by one weekly to adjust the dosage of drugs for 6 weeks.At the end of the 9 th week,rats in each group were sacrificed.Blood samples were collected and serum was prepared routinely to detect the contents of blood lipids[total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-G)],free fatty acids(FFA)and nitric oxide(NO).The contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in liver homogenate were also determined.The histopathological examination of liver tissues was conducted to observe the degree of steatosis in liver cells by hematoxylin and eosin(HE)staining.Results:1)Pathological pictures showed that the volume of liver cells in the model group increased,and mixed lipid droplets with inflammatory cell infiltration were seen in the cytoplasm of the cells.QWTX reduced the content of liver lipids and inhibited the lipid deposition.2)In blood lipid levels,there was significant difference between QWTX group and model group(P<0.05).3)Compared with model group,QWTX groups decreased the levels of MDA in liver and FFA in serum,and increased the levels of SOD in liver and NO in serum(P<0.05).Conclusion:QWTX could improve the lipid metabolism of NAFLD,and the mechanism might be related to anti-lipid peroxidation.
