摘 要:为寻求一种以水为溶剂高效提取核桃青皮总黄酮的最优工艺,以便后续核桃青皮堆肥归田还林,实验采用超声波辅助复合酶提取核桃青皮总黄酮进行研究。首先通过单因素实验分析了复合酶配比、浸取温度、料液比、浸取时间和浸取液pH等因素对核桃青皮总黄酮提取率的影响,然后利用响应面分析法优化提取工艺条件,最后通过三组平行实验验证优化结果。实验表明:在功率为100 W的超声环境中,当复合酶配比(果胶酶/纤维素酶)为1∶1,浸取温度为55℃,液料比(g/m L)为1∶35,浸取时间为30 min,浸取液pH为6时,总黄酮提取率最高,约为7.53%。In order to find an optimal technology of extracting total flavonoids from walnut green husk efficiently with water as solvent,so as to return walnut green husk compost to farmland and forest,the extraction of total flavonoids from walnut green husk by ultrasonic-assisted compound enzyme method was studied in this experiment.Firstly,the influence of factors such as compound enzyme ratio,extraction temperature,material liquid ratio,extraction time and pH of extraction solution on the extraction rate of total flavonoids from walnut green husk was analyzed by single factor experiment.Then,the extraction process conditions were optimized by response surface analysis.Finally,the optimization results were verified by three groups of parallel experiments.The results showed that in the ultrasonic environment with power of 100 W,when the ratio of compound enzyme(pectinase/cellulase)was 1:1,the extraction temperature was 55℃,the ratio of liquid to material(g/mL)was 1:35,the extraction time was 30 min,and the pH of the extraction solution was 6,the extraction rate of total flavonoids was the highest,about 7.53%.
