摘　　要：目的探讨miR-1225-5p过表达对宫颈癌Hela细胞增殖、凋亡、侵袭和迁移的影响。方法qRT-PCR检测20例宫颈癌患者癌旁正常组织和癌组织及Etc1/E6E7、Caski、Hela、SiHa、C4-1细胞系中miR-1225-5p的表达。选择宫颈癌Hela细胞进行研究。用Lipofectamine 2000将miR-1225-5p mimic和miR-NC转染到Hela细胞。采用CCK-8、细胞克隆形成实验、流式细胞术、Transwell实验和细胞划痕实验分别检测Hela细胞的增殖、凋亡、侵袭和迁移;Western blot检测Bcl-2、Bax、cleaved caspase-3、cleaved caspase-9、E-cadherin、N-cadherin和Vimentin蛋白表达水平。结果宫颈癌组织中miR-1225-5p的表达水平低于癌旁正常组织(P<0.01),宫颈癌细胞系中miR-1225-5p的表达水平降低(P<0.05)。过表达miR-1225-5p后Hela细胞增殖能力、克隆形成率、侵袭细胞数和划痕愈合率降低(P<0.01),Hela细胞凋亡率升高(P<0.01),Bax/Bcl-2比值、cleaved caspase-3/caspase-3、cleaved caspase-9/caspase-9和E-cadherin蛋白表达水平上调(P<0.01),N-cadherin和Vimentin蛋白表达水平下调(P<0.01)。结论miR-1225-5p过表达能抑制Hela细胞增殖、侵袭和迁移,并诱导细胞凋亡。Objective To investigate the effects of overexpression of miR-1225-5p on the proliferation,apoptosis,invasion and migration of Hela cells of cervical cancer.Methods qRT-PCR was used to detect the expression of miR-1225-5p in adjacent normal tissues in 20 cervical cancer patients and cancer tissues and Etc1/E6E7,Caski,Hela,SiHa and C4-1 cell lines.The Hela cells of cervical cancer were selected for study.miR-1225-5p mimic and miR-NC were transfected into Hela cells with Lipofectamine 2000.CCK-8,cell clone formation assay,flow cytometry,Transwall assay and cell scratch wound healing assay were used to detect the proliferation,apoptosis,invasion and migration of Hela cells.The protein expression levels of Bcl-2,Bax,cleaved caspase-3,cleaved caspase-9,E-cadherin,N-cadherin and Vimentin were detected by Western blot.Results The expression level of miR-1225-5p in the cervical cancer tissues was lower than that in the adjacent normal tissues(P<0.01),and the expression level of miR-1225-5p in the cervical cancer cell lines was reduced(P<0.05).After overexpression of miR-1225-5p,the proliferation ability,colony formation rate,number of invasive cells and scratch wound healing rate of Hela cells decreased(P<0.01),the apoptosis rate of Hela cells increased(P<0.01),the Bax/Bcl-2 ratio,the expression levels of cleaved caspase-3/caspase-3,cleaved caspase-9/caspase-9 and E-cadherin protein were up-regulated(P<0.01),and the expression levels of N-cadherin and Vimentin protein were down-regulated(P<0.01).Conclusion Overexpression of miR-1225-5p can inhibit the proliferation,invasion and migration of Hela cells,and induce cell apoptosis.
