摘  要:目的 探讨miR-146a调控Notch受体2(NOTCH2)表达对剥脱综合征(XFS)性青光眼患者炎性因子白细胞介素6(IL-6)水平的影响。方法 选取15例XFS性青光眼患者为XFS性青光眼组,15例正常健康人群为对照组。采用实时荧光定量PCR检测两组研究对象外周血miR-146a、NOTCH2 mRNA表达水平;采用Western blot检测两组研究对象外周血NOTCH2蛋白表达水平;采用ELISA检测两组研究对象血清IL-6表达水平。结果 XFS性青光眼组的miR-146a表达水平低于对照组,NOTCH2 mRNA表达水平和蛋白表达水平、血清IL-6表达水平高于对照组(均P<0.05)。结论 XFS性青光眼患者miR-146a、NOTCH2、IL-6表达异常。miR-146a可能通过靶向调控NOTCH2基因及蛋白表达水平作用于炎性因子IL-6,引发炎性反应,从而参与XFS性青光眼的发生和发展过程。Objective To investigate the effect of miR-146 a regulating Notch receptor 2(NOTCH2) expression on inflammatory factor interleukin 6(IL-6) level in glaucoma patients with exfoliation syndrome(XFS).Methods Fifteen glaucoma patients with XFS were selected as the XFS glaucoma group,and 15 normal healthy people as the control group.The expression levels of miR-146 a and NOTCH2 mRNA in peripheral blood of the two groups were detected by the real-time fluorescent quantitative PCR,and the Western blot was used to detect the expression of NOTCH2 protein in peripheral blood of the two groups;in addition,the ELISA was performed to detect the expression of serum IL-6 in the two groups.Results The expression of miR-146 a in the XFS glaucoma group was lower than that in the control group,and the expressions of NOTCH2 mRNA and protein,as well as serum IL-6 expression were higher in the XFS glaucoma group than in the control group(all P<0.05).Conclusion The expressions of miR-146 a,NOTCH2 and IL-6 are abnormal in glaucoma patients with XFS.MiR-146 a may be involved in the occurrence and development of XFS glaucoma through targetedly regulating the expressions of NOTCH2 gene and protein then acting on the inflammatory factor IL-6 to cause inflammatory response.
