摘  要:目的探索采用“饮食失节+过度疲劳”法建立脾虚肌萎证小鼠模型的可能性。方法昆明小鼠采用“饮食失节+过度疲劳”法建立脾虚肌萎证小鼠模型,观测并记录各组小鼠的外观行为评分、体质量及饮食量、负重游泳力竭时间、抓握力、胸腺与脾脏指数、肌湿重与肌重/体质量比;Elisa法测定小鼠血清D-木糖、胃泌素(gastrin,GAS)、胃动素(motilin,MTL)、血管活性肠肽(vasoactive intestinal peptide,VIP)、白细胞介素(interleukin,IL)-1β、IL-6、肿瘤坏死因子(tumor necrosis factor,TNF-α),观察病理改变并计算肌纤维直径与横截面面积。结果与正常组比较,模型小鼠第2~4周活动状态、皮肤毛发、大便状态积分均明显增高(P<0.01),第1~4周体质量与抓握力明显降低(P<0.01),第3~4周饮食量明显降低(P<0.01),第2~4周游泳力竭时间明显缩短(P<0.05),第2、4周血清D-木糖、GAS、MTL含量明显降低,VIP、IL-6含量明显增加(P<0.05),IL-1β、TNF-α含量与胸腺、脾脏指数明显降低(P<0.05),腓肠肌湿重和湿重/体质量比明显降低(P<0.05),光镜下的腓肠肌纤维排列松散,边缘变钝,肌纤维间隙与肌束间隙明显变宽,肌纤维直径与横截面面积明显减少(P<0.05)。结论“饮食失节+疲劳过度”法建立的模型小鼠在呈现中医脾虚证候相关的外观行为和实验室指标改变的同时,伴有骨骼肌肌力下降与肌纤维萎缩,表明该法可成功建立小鼠脾虚肌萎证模型。Objective:Exploring the possibility of establishing a mouse model of muscular dystrophy with spleen deficiency by the method of"food control+exhausted swimming".Methods:KM mice adopted the"food control+exhausted swimming"method to establish a spleen-deficiency and muscle weakness mouse syndrome model.The appearance and behavior scores,body weight and diet,weight-bearing swimming exhaustion time,grasping power,thymus and spleen index,wet muscle weight and muscle weight/body weight ratio of mice in each group were observed and recorded.The Elisa method was used to measure serum D-xylose,GAS,MTL,VIP,IL-1β,IL-6,TNF-α.Pathological changes were observed,and the muscle fiber diameter and cross-sectional area were calculated.Results:Compared with the control group,mice in the model group,the scores of activity status,skin and hair,and stool status were increased significantly in the 2~4 weeks of the experiment(P<0.01),the weight and grasping power were significantly reduced in the 1~4 weeks(P<0.01),the food intake in the 3~4 weeks was decreased significantly(P<0.01),the swimming exhaustion time was significantly shortened in the 2~4 weeks(P<0.05).In the 2^(nd) and 4^(th) weeks,mice in the model group,the levels of D-xylose,GAS,MTL in serum were significantly decreased,and the levels of VIP,IL-6 in serum were significantly increased(P<0.05),the levels of IL-1βand TNF-αin serum and the thymus and spleen indexes were significantly reduced(P<0.05),the wet weight and wet weight/body weight ratio of gastrocnemius muscle were significantly reduced(P<0.05).Observed by light microscope,mice in the model group showed that the gastrocnemius muscle fibers were arranged loosely,the edges became blunt,the muscle fiber gap and the muscle bundle gap were obviously widened,and the muscle fiber diameter and cross-sectional area were significantly reduced(P<0.05).Conclusions:The mouse model established by the"food control+exhausted swimming"method not only showed the appearance and behavior related to the syndrome of spleen deficiency
