摘 要:目的:探讨牛膝甾酮对成牙骨质细胞OCCM-30成骨效应及Notch信号通路的影响。方法:根据牛膝甾酮作用剂量的不同将OCCM-30细胞分为空白对照组(0mg/L)、牛膝甾酮低剂量组(2.5mg/L)、牛膝甾酮中剂量组(5.0mg/L)、牛膝甾酮高剂量组(10.0mg/L)。CCK-8法检测细胞增殖水平,Fluo-3 AM探针检测细胞内钙离子荧光强度,流式细胞术检测细胞凋亡率,RT-qPCR检测Notch1、DSL、CSL mRNA表达,Western Bloting检测Notch1、DSL、CSL蛋白表达。结果:与空白对照组比较,牛膝甾酮低、中、高剂量组OCCM-30细胞增殖率、钙离子荧光强度、Notch1、DSL、CSL mRNA及蛋白表达水平均显著升高,OCCM-30细胞凋亡率显著降低,且牛膝甾酮各剂量组OCCM-30细胞增殖率、钙离子荧光强度、Notch1、DSL、CSL mRNA及蛋白表达水平呈低剂量组<中剂量组<高剂量组,牛膝甾酮各剂量组OCCM-30细胞凋亡率呈低剂量组>中剂量组>高剂量组,差异均有统计学意义(P<0.05)。结论:膝甾酮可激活Notch信号通路相关蛋白表达,从而促进成牙骨质细胞OCCM-30成骨效应。Objective:To investigate the effect of achyosterone on the osteogenesis of cementoblasts OCCM-30 and the Notch signaling pathway.Method:OCCM-30 cells were divided into blank control group(0mg/L),low-dose group(2.5mg/L),medium-dose group(5.0mg/L)and the high-dose group of achyosterone(10.0mg/L)according to the different doses of achyranone.The level of cell proliferation was detected by CCK-8 method,the intracellular calcium ion detected by Fluo-3 AM probe,the apoptosis rate was detected by flow cytometry,the mRNA expression of Notch1,DSL and CSL was detected by RT-qPCR,and Notch1,DSL,and CSL protein expression were detected by western blotting.Results:Compared with the blank control group,the proliferation rate,calcium ion fluorescence intensity,Notch1,DSL,CSL mRNA and protein expression levels of OCCM-30 cells in each dose group of achyosterone were significantly increased,the apoptosis rate of OCCM-30 was decreased,and the cell proliferation rate,calcium ion fluorescence intensity,Notch1,DSL,CSL mRNA and protein expression levels of OCCM-30,and apoptosis rate in each dose group of achyosterone were dose-dependent,and the differences were statistically significant(P<0.05).Conclusion:Achyosterone can activate the expression of related proteins of Notch signaling and promot the osteogenic effect of cementoblast OCCM-30.
