摘 要:目的:从首乌叶中分离纯化得到杨梅苷、槲皮苷及阿福豆苷3个黄酮苷,并建立HPLC法同时测定首乌叶中这3个黄酮苷的含量。方法:利用MCI中高压色谱及半制备液相色谱进行分离纯化,HPLC检测采用SHIMADZU Shimpack GIST C18色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈(A)-水(B),梯度洗脱;柱温35℃;流速1.0 mL/min;检测波长350 nm;进样量10μL。结果:从首乌叶中首次分离纯化得到3个黄酮类化合物,鉴定为杨梅苷(1)、槲皮苷(2)和阿福豆苷(3)。3种成分在各自范围内线性关系良好(R^(2)>0.9990);平均加样回收率为97.29%~99.49%,RSD为1.26%~1.91%。结论:该方法操作简单、方便易行、重复性高、专属性强,可用于首乌叶中3个黄酮苷的含量测定。Objective:To separate three flavonoid glycosides composed of myricitrin,quercitrin and afzelin and establish a method of simultaneous determination of these three flavonoid glycosides in the leaves of Polygonum multiflorum Thunb.by HPLC.Methods:MCI chromatograph system and semi-preparative HPLC are used to separate the sample.The conditions of HPLC are as follows:the column is SHIMADZU Shimpack GIST C18 column and the mobile phase were consisted of acetonitrile(A)and water(B)with the gradient elution.The column temperature is 35℃and the detection wavelength is 350 nm;the injection volume is 10μL and the flow rate is 1.0 mL/min.Results:Three flavonoids are isolated for the first time from the PML,which are identified as myricitrin(1),quercitrin(2)and afzelin(3).Three constituents exhibited good linear relationships within their own ranges(R^(2)>0.9990),whose average recoveries are 97.29%~99.49%,the RSD is 1.26%~1.91%,respectively.Conclusion:The method in this study is appropriately used in the determination of three flavonoid glycosides in PML with good specificity,simplicity and high repeatability.
