摘  要:目的利用在线生物信息数据库,组织和细胞水平实验探究SIM2基因在膀胱癌中的表达及涉及的临床意义。方法通过UALCAN数据库分析SIM2基因在膀胱癌患者和正常膀胱组织中的表达差异,SIM2基因表达与膀胱癌患者淋巴结转移、分期、组织亚型等临床病理参数的关系;GEPIA数据库、HPA数据库、组织芯片和定量PCR技术验证SIM2基因在膀胱癌患者中的mRNA和蛋白表达差异;利用String数据库分析SIM2基因的蛋白相互作用及调控网络;Linked Omics数据库做SIM2相关分子的富集分析;TIMER和TISIDB数据库分析SIM2基因与肿瘤微环境免疫细胞浸润的关系。结果UALCAN、GEPIA和HPA数据库结果显示,膀胱癌组织中SIM2基因的mRNA和蛋白水平的表达明显高于正常膀胱组织(P<0.05);UALCAN在线工具分析揭示,SIM2的表达与患者的淋巴结转移、病理类型、分期密切相关;免疫组织化学及定量PCR湿实验结果显示,SIM2蛋白和mRNA表达水平在膀胱癌组织中明显升高(P<0.05),但SIM2的表达与膀胱癌患者TNM分期和病理分级无相关性;String在线分析结果提示,与SIM2具有相互作用的蛋白涉及生物过程包括参与类泛素蛋白修饰、转录调控、蛋白水解等生物学过程;LinkedOmics富集分析表明,与SIM2正相关基因涉及TNF、NF-kappa B和JAK-STA等信号通路及多种生物学功能,包括细胞黏附分子结合,G蛋白偶联受体结合和细胞因子受体活性等;免疫细胞浸润水平显示SIM2的表达与树突状细胞等呈负相关。结论通过在多种在线数据库及实验结果揭示SIM2基因在膀胱癌中高表达,可能为膀胱癌发生发展的重要分子靶点。Objective To explore the expression and clinical significance of SIM2 gene in bladder cancer through bioinformatics analysis,tissues and cell tests.Methods UALCAN database was performed to analyze the different expression of SIM2 gene in bladder cancer tissues and normal bladder tissues,and the association between SIM2 gene expression and clinicopathological parameters including lymph node metastasis,stage,tissue subtype of bladder cancer patients were further examined.GEPIA database,HPA database and tissue chip were used to evaluate the mRNA and protein expression levels of SIM2 in bladder cancer patients.Furthermore,String database was applied to construct protein-protein interaction(PPI)network of SIM2 coding protein.LinkedOmics was used for function enrichment analysis of SIM2.The relationship between SIM2 and tumor lymphocyte infiltration was analyzed in TIMER and TISIDB database.Results UALCAN,GEPIA and HPA database showed that the mRNA and protein levels of SIM2 gene in bladder cancer tissues were obviously higher than those in normal bladder tissues(P<0.05).UALCAN database analysis revealed that the expression of SIM2 was closely related to lymph node metastasis,histopathological type and stage of patients.The results of immunohistochemical showed that SIM2 protein expression level was significantly increased in bladder cancer tissues(P<0.05),and which was not correlated with TNM stage or pathological grade of bladder cancer patients.PPI network suggested that SIM2 protein was closely associated with biological processes,including ubiquitin-like protein modification,transcriptional regulation,and protein hydrolysate.The enrichment analysis of LinkedOmics showed that SIM2 positively correlated genes involved in TNF,NF-KappaB and JAK-STAT signaling pathways,and a variety of biological functions,containing cell adhesion function,G protein coupled receptor function,cytokine receptor activity.The results of immune cell infiltration showed that SIM2 expression was negatively correlated with dendritic cells.Conc
