EZH2抑制剂(GSK-126)对小鼠肝纤维化的保护作用

摘　　要：观察EZH2抑制剂(GSK-126)对小鼠肝纤维化的保护作用.将24只实验小鼠随机分为4个小组:对照组(食用油注射量:5 mL/kg)、模型组(CCl_(4)注射量:5 mL/kg的20%的橄榄油溶液)、CCl_(4)+GSK-1265 mg/kg组(CCl_(4)注射量:5 mL/kg的20%的橄榄油溶液,GSK-126注射量:5 mg/kg)、CCl_(4)+GSK-12615 mg/kg组(CCl_(4)注射量:5 mL/kg的20%的橄榄油溶液,GSK-126注射量:15 mg/kg).每3天注射1次,20天后解剖,观察肝脏形态、病理情况及血清ALT值,并通过HE染色和天狼猩红染色来检测GSK-126对小鼠肝纤维化的影响.结果发现:两个GSK-126处理组的肝脏质地较软、肝重指数较轻、ALT活性较低、血管交联和胶原蛋白水平较低,且GSK-12615 mg/kg组比GSK-1265 mg/kg组的肝纤维化程度更低(p<0.05),说明GSK-126对肝纤维化小鼠有明显的保护作用.In order to observe the preventive effect of EZH2 inhibitor(GSK-126)on liver fibrosis in mice,24 mice were randomly divided into four groups:the control group(with oil injection:5mL/kg),the model group(with CCl_(4) injection:20%olive oil solution,5 mL/kg),the CCl_(4)+GSK-126 low-dose group(CCl_(4) injection:20%olive oil solution,5 mL/kg,GSK-126 injection:5 mg/kg),and the CCl_(4)+GSK-126 high-dose group(CCl_(4) injection:20%olive oil solution,5 mL/kg,GSK-126 injection:15 mg/kg).The injection was taken once every three days.Twenty days later,the mice were dissected to observe the liver morphology and pathology,measure the serum ALT and detect the effect of GSK-126 on liver fibrosis by HE staining and Sirius red staining.The results showed that the livers of the two GSK-126-treated groups were softer and had lower hepatosomatic index,ALT activity,vascular adhesion and collagen level,and the liver fibrosis degree of the high-dose group was lower than that of the low-dose group(p<0.05),which demonstrates that GSK-126 has a preventive effect on hepatic fibrosis in mice.
