摘  要:目的:基于Catwalk步态分析系统,评价三化汤对大脑中动脉闭塞(MCAO)大鼠运动功能缺损的改善效果。方法:将36只成年雄性Wistar大鼠随机分为模型组、三化汤低剂量组、三化汤中剂量组、三化汤高剂量组、尼莫地平组、假手术组,每组6只,前5组采用Zea Longa线栓法构建MCAO大鼠模型,假手术组大鼠接受同样的手术,只是将栓线向前推进到颈内动脉约5 mm处。4个药物组分别给予对应药物灌胃,假手术组与模型组灌胃等体积生理盐水,每日灌胃1次,共7 d。造模前后分别进行Zea Longa神经功能缺损评分,灌胃结束后进行Catwalk步态数据采集。结果:造模后第1天,与假手术组比较,5个造模组Zea Longa神经功能缺损评分均显著升高(P<0.01);造模第7天,三化汤高剂量组Zea Longa神经功能缺损评分低于模型组(P<0.01)。治疗后,三化汤低、中、高剂量组和尼莫地平组平均运动速度、持续时间、站立时间、摆动时间、摆动速度、站立行走模式、步幅、爪间宽度均显著改善,且三化汤各剂量组效果优于尼莫地平组(P<0.05、P<0.01)。结论:三化汤可以增强MCAO大鼠的运动能力,改善神经功能缺损症状,效果优于尼莫地平。Objective:Based on the Catwalk gait analysis system,to evaluate the improvement effect of Sanhua Decoction(三化汤)on motor deficit in rats with middle cerebral artery occlusion(MCAO).Methods:Thirty six adult male Wistar rats were randomly divided into model group,Sanhua Decoction low dose group,Sanhua Decoction medium dose group,Sanhua Decoction high dose group,nimodipine group and sham operation group,with 6 rats in each group.The first five groups were used the Zea Longa suture method to construct the MCAO rat model.The rats in the sham operation group received the same operation,but the suture was advanced to the internal carotid artery about 5 mm.The 4 drug groups were respectively given the corresponding drugs by gavage,and the sham operation group and the model group were given the same volume of normal saline by gavage,once a day,for a total of 7 days.The Zea Longa neurological deficit scores were performed before and after modeling,and the Catwalk gait data was collected after gavage.Results:On the first day after modeling,compared with the sham operation group,the Zea Longa neural function defect scores of the five modeling groups were significantly higher(P<0.01);on the 7th day after modeling,the Zea Longa neural function defect score in the high dose group of Sanhua Decoction was lower than that in the model group(P<0.01).After treatment,average movement speed,duration,standing time,swing time,swing speed,standing walking pattern,stride length,and interpaw width were significantly improved in the low,medium,and high dose groups of Sanhua Decoction and the nimodipine group,and the effect of each dose group of Sanhua Decoction was better than that of nimodipine group(P<0.05,P<0.01).Conclusion:Sanhua Decoction can enhance the motor ability of MCAO rats and improve the symptoms of neurological deficit,and the effect is better than that of nimodipine.
