SOCS基因家族在肝细胞癌中的表达及生物信息学分析

摘　　要：目的基于高通量生物信息学方法系统分析细胞因子信号抑制物(suppressor of cytokine signaling,SOCS)基因家族在肝细胞癌(hepatocellular carcinoma,HCC)发生、发展中的作用机制。方法使用Oncomine、UNLCAN、cBioPortal、Kaplan-Meier plotter、David等多个在线数据库对SOCS家族分子在肝癌细胞中的表达差异、临床病理特征、生存分析、基因突变、生物学功能方面进行综合分析。String在线数据库用于构建SOCS家族基因的蛋白质-蛋白质相互作用网络。结果大部分SOCS家族基因在HCC中存在表达差异,SOCS5和SOCS7表达显著上调(P<0.05),而SOCS2、SOCS3、SOCS6、CISH表达下调(P<0.05)。该家族基因SOCS1、SOCS2、SOCS4、SOCS5、SOCS7、SOCS8与预后相关(P<0.05)。SOCSs家族成员在HCC患者中的基因改变形式主要包括扩增、突变和多重基因等类型。在HCC与SOCS家族相关的生物学过程主要富集在Jak-STAT、炎症反应等方面。结论SOCS基因家族大部分成员在HCC中差异表达,可能是治疗肝细胞癌潜在的新靶点。Objective:To systematically analyze the mechanism of suppressor of cytokine signaling(SOCS)gene family in hepatocellular carcinoma(HCC)based on high-throughput bioinformatics method.Methods:Multiple online databases,such as Oncomine,UNLCAN,cBioPortal,Kaplan-Meier plotter,David,etc.were used to comprehensively analyze the expression differences,clinicopathological characteristics,survival analysis,gene mutations,and biological functions of SOCS family molecules in hepatocellular carcinoma.The String online database was used to construct the protein-protein interaction network of SOCS family genes.Results:Most SOCS family genes were differentially expressed in HCC.The expressions of SOCS5 and SOCS7 were significantly up-regulated(P<0.05),while the expressions of SOCS2,SOCS3,SOCS6 and CISH were down-regulated(P<0.05).The family gene prognosis analysis SOCS1,SOCS2,SOCS4,SOCS5,SOCS7,SOCS8 were associated with prognosis(P<0.05).The gene alteration forms of SOCS family members in HCC patients mainly include amplification,mutation and multiple gene types.The biological processes related to the SOCS family in HCC are mainly enriched in Jak-STAT,inflammatory response and other aspects.Conclusion:Most members of the SOCS gene family are differentially expressed in HCC,and may be potential new targets and prognosis-related genes for the treatment of hepatocellular carcinoma in the future.
