膀胱尿路上皮癌组织中LncRNA NNT-AS1、miR-22表达与预后的关系

摘　　要：目的分析长链非编码RNA(LncRNA)烟酰胺核苷酸反义转氢酶RNA1(NNT-AS1)及微小RNA-22(miR-22)在膀胱尿路上皮癌(BUC)组织中的表达以及二者与患者预后的关系。方法选取本院收治的BUC患者39例,留取患者术中切除的癌组织及癌旁正常组织;另选取人正常膀胱上皮细胞SV-HUC-1作为对照组,人膀胱癌细胞株T24作为BUC细胞组。采用qRT-PCR法检测细胞及组织中LncRNA NNT-AS1、miR-22的表达水平,分析二者与患者临床病理特征的关系;采用Pearson法分析癌组织中LncRNA NNT-AS1、miR-22的相关性,Kaplan-Meier法评估LncRNA NNT-AS1、miR-22表达与BUC患者总生存率的关系。结果BUC细胞组中LncRNA NNT-AS1水平高于对照组(P<0.05),miR-22水平低于对照组(P<0.05)。BUC患者癌组织中LncRNA NNT-AS1水平高于癌旁正常组织(P<0.05),miR-22水平低于癌旁正常组织(P<0.05)。LncRNA NNT-AS1、miR-22水平与患者肿瘤浸润、病理分级及淋巴结转移有关(P<0.05),且miR-22水平与患者临床分期有关(P<0.05)。癌组织中LncRNA NNT-AS1水平与miR-22水平呈负相关(P<0.05)。LncRNA NNT-AS1高表达组总生存率低于LncRNA NNT-AS1低表达组(P<0.05);miR-22高表达组总生存率高于miR-22低表达组(P<0.05)。LncRNA NNT-AS1高表达+miR-22低表达患者总生存率低于LncRNA NNT-AS1低表达+miR-22高表达患者及其他患者,3组比较差异有统计学意义(P<0.05)。结论LncRNA NNT-AS1、miR-22在BUC细胞及癌组织中异常表达,二者呈负相关,且均与患者临床病理特征及预后有关。Objective To analyze the expression of long non-coding RNA(LncRNA)nicotinamide nucleotide transhydrogenase-antisense RNA1(NNT-AS1)and microRNA-22(miR-22)in bladder urothelial carcinoma(BUC)tissue and their relationship with prognosis.Methods A total of 39 BUC patients admitted to this hospital were selected,and the cancer tissues and adjacent normal tissues removed during the operation were collected.Human normal bladder epithelial cell SV-HUC-1 was selected as the control group and human bladder cancer cell line T24 was selected as the BUC cell group.The qRT-PCR method was used to detect the expression levels of LncRNA NNT-AS1 and miR-22 in the cells and tissues,and the relationship between the two and the clinical pathological characteristics of the patients were analyzed.Pearson method was used to analyze the correlation of LncRNA NNT-AS1 and miR-22 in cancer tissues,and Kaplan-Meier method was used to evaluate the relationship between the expression of LncRNA NNT-AS1 and miR-22 and the overall survival rate of BUC patients.Results The level of LncRNA NNT-AS1 in BUC cell group was higher than that in control group(P<0.05),and the level of miR-22 was lower than that in control group(P<0.05).The level of LncRNA NNT-AS1 in cancer tissues of BUC patients was higher than that of normal tissues adjacent to cancer(P<0.05),and the level of miR-22 was lower than that of normal tissues adjacent to cancer(P<0.05).The levels of LncRNA NNT-AS1 and miR-22 were related to tumor invasion,pathological grade and lymph node metastasis(P<0.05),and the level of miR-22 was related to clinical stage(P<0.05).The level of LncRNA NNT-AS1 in cancer tissues was negatively correlated with the level of miR-22(P<0.05).The overall survival rate of LncRNA NNT-AS1 high expression group was lower than that of LncRNA NNT-AS1 low expression group(P<0.05),and the overall survival rate of miR-22 high expression group was higher than that of miR-22 low expression group(P<0.05).The overall survival rate of patients with high expression of LncRNA NNT-
