摘 要:目的探讨骨肉瘤干细胞生物标志物CD117与Stro-1的表达及其临床意义。方法选取58例骨肉瘤患者的手术切除标本作为骨肉瘤组,另取其中13例患者的瘤旁正常骨组织标本作为骨组织组。取2组组织蜡片构建组织芯片,运用免疫组织化学法检测骨肉瘤组与骨组织组中骨肉瘤干细胞生物标志物CD117与Stro-1的表达,同时分析CD117、Stro-1表达与骨肉瘤患者临床病理因素的关系。结果骨肉瘤组的CD117与Stro-1阳性表达率[72.41%(42/58)、68.97%(40/58)]高于骨组织组[30.77%(4/13)、30.77%(4/13)],有统计学差异(P<0.05)。不同年龄、不同性别、不同肿瘤直径中CD117、Stro-1表达水平比较,无统计学差异(P>0.05);不同组织学类型、是否发生肿瘤转移、TNM各分期中CD117、Stro-1表达水平比较,有统计学差异(P<0.05)。结论骨肉瘤组织中干细胞生物标志物CD117与Stro-1的阳性表达均处于较高水平,且与组织学类型、肿瘤转移、TNM分期相关,CD117、Stro-1有望作为判断骨肉瘤恶性程度的重要参考指标。Objective To investigate the expression and clinical significance of stem cell biomarkers CD117 and STRO-1 in osteosarcoma.Methods 58 patients with osteosarcoma resected and confirmed by pathological examination were divided into osteosarcoma group,and 13 of them were divided into bone tissue group.Take 2 groups of tissue wax to construct tissue microarray,and detect the expression of osteosarcoma stem cell biomarkers CD117 and STRO-1 in osteosarcoma group and bone tissue group by immunohistochemistry.At the same time,analyze the relationship between the expression of CD117 and STRO-1 and the clinicopathological factors of osteosarcoma patients.Results The positive expression rates of CD117 and STRO-1 in osteosarcoma Group[72.41%(42/58),68.97%(40/58)]were significantly higher than those in bone tissue group[30.77%(4/13),30.77%(4/13)](P<0.05);There was no significant difference in the expression levels of CD117 and STRO-1 in different age,gender and tumor diameter(P>0.05);There were significant differences in the expression levels of CD117 and STRO-1 in different histological types,tumor metastasis and TNM stages(P<0.05).Conclusion The positive expression of stem cell biomarkers CD117 and STRO-1 in osteosarcoma is at a high level,which is related to histological type,tumor metastasis and TNM stage.CD117 and STRO-1 are expected to be important reference indexes to judge the malignancy of osteosarcoma.
