摘 要:目的 探讨肺腺癌转移相关转录本1(MALAT1)对神经病理性疼痛(NP)大鼠信号转导和转录激活因子3(STAT3)信号通路的影响。方法 选取置管成功的大鼠40只,随机分为对照组(注射生理盐水)、假手术组(仅暴露坐骨神经+注射生理盐水)、NP组(NP造模+注射生理盐水)、阴性慢病毒组(NP造模+注射阴性慢病毒干扰载体)和MALAT1 shRNA组(NP造模+注射MALAT1慢病毒干扰载体)。测定大鼠机械性缩足反射阈值(MWT)和热缩足反射潜伏期(TWL),采用实时荧光定量聚合酶链式反应(qRT-PCR)法检测大鼠脊髓组织中MALAT1、STAT3 mRNA表达情况,采用酶联免疫吸附试验(ELISA)法检测大鼠脊髓组织中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)表达水平,采用蛋白免疫印迹(Western Blot)法检测大鼠脊髓组织中磷酸化STAT3(p-STAT3)、STAT3蛋白表达情况。结果 与假手术组比较,造模后NP组大鼠MWT明显降低(P<0.05),TWL明显缩短(P<0.05),大鼠脊髓组织中MALAT1、STAT3 mRNA表达水平,IL-6、TNF-α、IL-1β表达水平及p-STAT3/STAT3蛋白表达水平明显升高(P<0.05);与阴性慢病毒组比较,MALAT1 shRNA组大鼠MWT明显升高(P<0.05),TWL明显延长(P<0.05),大鼠脊髓组织中MALAT1、STAT3 mRNA表达水平,IL-6、TNF-α、IL-1β表达水平及p-STAT3/STAT3蛋白表达水平明显降低(P<0.05)。结论 干扰MALAT1能抑制炎症反应及STAT3信号通路的激活,缓解NP大鼠的机械痛觉过敏及热痛觉过敏症状。Objective To investigate the effects of metastasis associated in lung adenocarcinoma transcript 1(MALAT1) on neuropathic pain(NP) rats signal transducer and activator of transcription 3(STAT3) signaling pathway.Methods Forty SD rats were divided into control group(injection of normal saline),sham operation group(exposure of sciatic nerve only+injection of normal saline),NP group(modeling of NP+injection of normal saline),and negative lentivirus group(modeling of NP+injection of negative lentiviral interference vector),and MALAT1 shRNA group(modeling of NP+injection of MALAT1 lentiviral interference vector).The mechanical withdrawal threshold(MWT) and thermal withdrawal latency(TWL) of rats were measured.The expression of MALAT1 and STAT3 mRNA in the spinal cord tissue of rats was detected by real-time fluorescent quantitative PCR(qRT-PCR) method.The expression levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and interleukin-1β(IL-1β) in the spinal cord tissue of rats were detected by enzyme-linked immunoassay(ELISA).The expression of p-STAT3 and STAT3 proteins in rat spinal cord tissue was detected by Western Blot method.Results Compared with sham operation group, the MWT of NP group was significantly reduced(P<0.05),the TWL was significantly shortened(P<0.05),the expression levels of MALAT1 and STAT3 mRNAs, expression levels of IL-6,TNF-α and IL-1β,protein expression level of p-STAT3/STAT3 in the rat spinal cord tissue were significantly increased(P<0.05).Compared with negative lentivirus group, the MWT of rats in the MALAT1 shRNA group was significantly increased(P<0.05),and the TWL was significantly prolonged(P<0.05),the expression levels of MALAT1 and STAT3 mRNAs, expression levels of IL-6,TNF-α,and IL-1β,protein expression level of p-STAT3/STAT3 in the rat spinal cord tissue were significantly reduced(P<0.05).Conclusion Interfering with MALAT1 can inhibit the inflammatory response and the activation of STAT3 signaling pathway, and alleviate the symptoms of mechanical hyperalgesia and ther
