摘 要:目的探讨多形性腺瘤基因1(PLAG1)在结直肠癌(CRC)组织中的表达情况,分析其表达水平与CRC患者临床病理特征及预后的关系。方法选择2014年1月—2017年3月手术切除的CRC组织及相应癌旁组织80例,检测PLAG1在癌组织及相应癌旁组织中的表达水平。应用GEPIA2数据库及临床样本评估PLAG1表达水平与CRC患者临床病理特征和预后的相关性。结果CRC组织中PLAG1阳性表达率高于癌旁组织(P<0.01)。CRC组织中PLAG1的高表达与T分期、N分期和临床病理分期显著相关(P<0.05)。GEPIA2分析和临床样本Kaplan-Meier分析显示,PLAG1高表达CRC患者的总生存期和无病生存期均短于PLAG1低表达者(P<0.05);Cox回归分析提示PLAG1高表达是CRC患者预后独立危险因素(P<0.05)。KEGG通路富集分析结果显示,PLAG1基因可能参与调节WNT、Hedgehog、Notch、Hippo、mTOR、PI3K-Akt等多种信号通路。结论PLAG1在CRC组织中表达水平升高,且与T分期、N分期、临床病理分期和临床预后相关,是CRC预后的独立危险因素。Objective To investigate the expression of pleomorphic adenoma gene 1(PLAG1)in colorectal cancer(CRC)tissue,and to analyze the relationship between its expression level and the clinicopathological characteristics and prognosis of CRC patients.Methods Eighty cases of CRC tissues and corresponding paracancerous tissues surgically resected from January 2014 to March 2017 were selected to detect the expression level of PLAG1 in cancer tissues and corresponding paracancerous tissues.The GEPIA2 database and statistical analysis of clinical samples were used to evaluate the correlation between the expression level of PLAG1 and the clinicopathological characteristics and prognosis of CRC patients.Results The positive expression rate of PLAG1 in CRC tissues was higher than that in paracancerous tissues(P<0.01).The high expression of PLAG1 in CRC tissue was significantly correlated with T stage,N stage and clinicopathological stage(P<0.05).GEPIA2 analysis and Kaplan-Meier analysis of clinical samples showed that the overall survival and disease-free survival of CRC patients with high PLAG1 expression were shorter than those with low PLAG1 expression(P<0.05).Cox regression analysis indicated that high expression of PLAG1 was an independent risk factor for the prognosis of CRC patients(P<0.05).The results of KEGG pathway enrichment analysis showed that PLAG1 gene might be involved in the regulation of WNT,Hedgehog,Notch,Hippo,mTOR,PI3K-Akt and other signaling pathways.Conclusion The expression level of PLAG1 was up-regulated in CRC tissues,and correlated with T stage,N stage,clinicopathological stage and clinical prognosis of CRC.Therefore,PLAG1 can be used as an independent risk factor for the prognosis of CRC.
