摘 要:舍雷肽酶具有出色的酪蛋白水解和溶纤活性,临床被应用于多种疾病的治疗或辅助治疗。为了提高黏质沙雷氏菌(Serratia marcescens)发酵产舍雷肽酶的活力,优化了产酶培养基组成及其初始pH和培养时间。首先,通过单因素实验确定了发酵培养基的主要成分;然后,采用响应面分析法优化了麦芽浸粉、酵母浸粉和牛肉膏的质量浓度;最后,考察培养基初始pH和培养时间对产酶活力的影响。实验结果表明:较佳的培养基组成为麦芽浸粉11.9 g/L、酵母浸粉11.3 g/L、牛肉膏6.45 g/L、MnSO_(4)1 g/L、ZnSO_(4)1 g/L、K_(2)HPO_(4)5 g/L和NaCl 5 g/L,初始pH 8.0;在30℃,200 r/min条件下培养36 h,舍雷肽酶活力达到1121 U/mL,较优化前的229.3 U/mL提高了3.89倍。优良的产酶培养基为发酵生产舍雷肽酶的工业化应用打下了基础。Serrapeptase has excellent activities of proteolysis and fibrinolysis,and is used for the therapy or adjuvant therapy of many diseases.In order to improve the fermentation activity of serrapeptase produced by Serratia marcescens,the medium composition,its initial pH and fermentation time were optimized.Firstly,the main components in fermentation medium were selected through single factor experiments;then,the concentration of malt extract,yeast extract and beef extract were optimized through response surface analysis;finally,the effects of initial pH of medium and fermentation time on the enzyme activity were investigated.The results showed that the optimal medium was composed of 11.9 g/L malt extract,11.3 g/L yeast extract,6.45 g/L beef extract,1 g/L MnSO_(4),1 g/L ZnSO_(4),5 g/L K_(2)HPO_(4),5 g/L NaCl,with the initial pH 8.0.The cultivation was carried at 30℃and 200 r/min for 36 h,and the activity of serrapeptase reached 1121 U/mL,which was 3.89 times higher than 229.3 U/mL before optimization.The optimized medium formula for the high activity of enzyme gives a foundation for the industrial fermentation of serrapeptase by S.marcescens.
