摘  要:文章以超声辅助低浓度碱溶解的丝素蛋白溶液为原料,通过碱性蛋白酶、木瓜蛋白酶、风味蛋白酶、胰蛋白酶、中性蛋白酶、菠萝蛋白酶等水解丝素蛋白,从中选出水解条件最好的蛋白酶,再进行水解条件的优化,得到丝素肽样品。并运用扫描电镜、粒径分布和红外光谱检测方法,测定丝素肽的形貌、结构和长度变化。结果表明,碱性蛋白酶水解的效果最好。因此,用碱性蛋白酶酶解丝素蛋白,又经过单因素实验、正交实验,证明了只有当底物浓度为2%、加酶量为10000 U/g、温度为55℃、酶解时间为150 min、pH为10时,丝素肽的水解度最高,为8.55%。In this paper,silk fibroin solution dissolved by ultrasound-assisted low-concentration alkali is used as the raw material.Silk fibroin is hydrolyzed by alkaline protease,papain,flavor protease,trypsin,neutral protease,bromelain and so on.The protease with the best hydrolysis conditions is selected,and then the hydrolysis conditions are optimized to obtain silk fibroin peptide samples.The changes of morphology,structure and length of silk fibroin peptide are determined by scanning electron microscope,particle size distribution and infrared spectroscopy.The results show that the effect of hydrolysis by alkaline protease is the best.Therefore,silk fibroin is hydrolyzed by alkaline protease.Through single factor experiment and orthogonal experiment,it is proved that the hydrolysis degree of silk fibroin peptide is the highest of 8.55%only when the substrate concentration is 2%,the addition amount of enzyme is 10000 U/g,the temperature is 55℃,the enzymatic hydrolysis time is 150 min and the pH is 10.
