摘　　要：NEK2是一种丝/苏氨酸蛋白激酶,是中心体正确复制、分离、成熟和建立双极纺锤体所必需的,它参与调控细胞周期过程,最初发现其与永生有丝分裂A(NIMA)激酶具有广泛结构相似性。在胃癌组织中,NEK2的表达与肿瘤的分化程度、淋巴结转移及TNM分期有关,说明NEK2可能与胃癌的恶性程度及预后有关。研究NEK2在胃癌细胞定位并促进胃癌恶性生长的作用机制,为胃癌的临床诊疗提供新的靶点,已成为胃癌免疫治疗的研究热点。本文就目前NEK2与胃癌发生发展的关系,以及NEK2在胃癌治疗中的作用机制的研究进展进行综述。NEK2 is a filament/threonine protein kinase,it is necessary for the correct replication,isolation,mature and bipolar spindle establishment of centrosomes,which is involved in the regulation of cell cycle,and was originally found to have broad structural similarities to fungal mold immortalized mitotic A(NIMA).In gastric cancer tissues,the NEK2 expression is related to the tumor differentiation degree,lymph node metastasis and TNM stage,which indicate that NEK2 may be related to the degree of malignancy and the prognosis of gastric cancer.The study on the localization of NEK2 in the gastric cancer cells and the mechanism promoting the malignant growth of gastric cancer provides a new target for the clinical diagnosis and treatment of gastric cancer,which has become a research hotspot of gastric cancer immunotherapy.This paper reviewed the study advances in the relationship between NEK2 and gastric cancer occurrence,and the action mechanism of NEK2 in the treatment of gastric cancer.
