摘  要:目的 探讨紧密连接蛋白4(CLDN4)在胰腺癌中的预后价值及机制。方法 运用基因表达谱交互式分析(GEPIA)、UALCAN、人类蛋白质表达数据库(HPA)、Kaplan-Meier绘图仪等数据库检索CLDN4的信息,分析CLDN4在胰腺癌组织中的表达水平、启动子甲基化状态、临床病理参数和生存分析并探讨相关机制。结果 CLDN4在胰腺癌组织中的表达水平高于正常胰腺组织,差异有统计学意义(P<0.05),其表达定位于细胞膜。不同病理分期的CLDN4基因表达水平不同(P<0.05),肿瘤病理分期越高,CLDN4基因表达水平越高。胰腺癌中CLDN4基因启动子区甲基化水平与正常胰腺组织比较,差异有统计学意义(P<0.05)。生存分析中,CLDN4的mRNA高表达患者的总生存时间短于低表达患者(P<0.05)。在String数据库中筛选出相互作用强度排位靠前的10个与CLDN4有相互作用的蛋白质,分别是OCLN、CLDN7、CDH1、TJP1、CLDN12、CLDN34、TJP2、TJP3、EPHA2及CRHL2。结论 CLDN4在胰腺癌组织中高表达,其表达水平与胰腺癌的预后显著相关,可作为潜在的胰腺癌预后生物标志物。Objective To investigate the prognostic value and mechanism of claudin 4(CLDN4) in pancreatic cancer.Methods The expression level and promoter methylation status of CLDN4 in pancreatic cancer tissues, clinicopathological parameters, and patient survival were analyzed by gene expression profiling interactive analysis(GEPIA),UALCAN,human protein atlas(HPA),and Kaplan-Meier plotter, etc.Results The expression level of CLDN4 in pancreatic cancer tissues was higher than that in normal pancreatic tissues(P<0.05),and the expression was localized on cell membrane.The expression level of CLDN4 gene was different in different pathological stages of tumor(P<0.05),a higher expression of CLDN4 gene corresponded to a higher pathological stage.The methylation level of the promoter region of CLDN4 gene in pancreatic cancer tissue was significantly different from that in normal pancreatic tissue(P<0.05).The survival analysis showed that the overall survival of patients with high expression of CLDN4 mRNA was significantly shorter than that of patients with low expression(P<0.05).The top 10 proteins interacting with CLDN4 were found in string database, which were OCLN,CLDN7,CDH1,TJP1,CLDN12,CLDN34,TJP2,TJP3,EPHA2 and CRHL2.Conclusion CLDN4 is highly expressed in pancreatic cancer, and the expression level is significantly correlated with the prognosis.Therefore, CLDN4 may be used as a potential biomarker for prognosis of pancreatic cancer.
