摘 要:目的 探讨沉默miR-222-3p对病毒感染相关的甲状腺癌细胞增殖、迁移的影响及作用机制。方法 将培养后的人甲状腺癌细胞系(TPC-1)分为空白对照组、过表达miR-222-3p组、沉默miR-222-3p组。应用实时荧光定量法检测各组细胞miR-222-3p表达;应用EdU实验检测甲状腺癌TPC-1细胞增殖情况;应用TUNEL法检测甲状腺癌TPC-1细胞凋亡情况;应用Transwell小室实验检测各组细胞侵袭情况;应用细胞划痕实验检测各组细胞迁移情况;Western blot检测各组p53、Bcl-2、Bax相对表达量。结果 过表达miR-222-3p组的miR-222-3p表达水平高于空白对照组、沉默miR-222-3p组(P<0.05);沉默miR-222-3p组的miR-222-3p表达水平低于空白对照组(P<0.05)。过表达miR-222-3p组的细胞增殖率、侵袭及迁移细胞数、p53与Bax蛋白的相对表达量均高于空白对照组、沉默miR-222-3p组(P<0.05);凋亡率、Bcl-2蛋白的相对表达量均低于空白对照组、沉默miR-222-3p组(P<0.05)。沉默miR-222-3p组的细胞增殖率、侵袭及迁移细胞数、p53与Bax蛋白的相对表达量均低于空白对照组;凋亡率、Bcl-2蛋白的相对表达量均高于空白对照组(P<0.05)。结论 沉默miR-222-3p能够抑制病毒感染相关甲状腺癌细胞增殖、迁移,其作用是通过调控p53、Bcl-2、Bax凋亡相关蛋白的表达来实现。OBJECTIVE To investigate the effect and mechanism of silencing miR-222-3 p on the proliferation and migration of virus-related thyroid cancer cells. METHODS The cultured human thyroid cancer cell line(TPC-1) was divided into the blank control group, mir-222-3 p overexpression group and mir-222-3 p silencing group. The expression level of mir-222-3 p was detected by RT-PCR. The proliferation of thyroid cancer TPC-1 cells was detected by EdU assay. TUNEL method was used to detect the apoptosis of thyroid cancer TPC-1 cells. Transwell chamber test was used to detect the invasion of cells in each group. Cell migration was detected by cell scratch test. The relative protein expression of p53, Bcl-2 and Bax in each group were detected by Western blot. RESULTS The expression level of miR-222-3 p in the miR-222-3 p overexpression group was significantly higher than that in the blank control group and the miR-222-3 p silencing group(P<0.05). The expression level of miR-222-3 p in the miR-222-3 p silencing group was significantly lower than that in the blank control group(P<0.05). The cell proliferation rate, the number of invasive and migrating cells, and the relative expressions of p53 and Bax proteins in the miR-222-3 p overexpression group were significantly higher than those in the blank control group and miR-222-3 p silenced group(P<0.05);the apoptosis rate and the relative expression of Bcl-2 protein were significantly lower than those in the blank control group and the miR-222-3 p silencing group(P<0.05). The cell proliferation rate, the number of invading and migrating cells, and the relative expression of p53 and Bax proteins in the miR-222-3 p silencing group were significantly lower than those in the blank control group, while the apoptosis rate and the relative expression of Bcl-2 protein were significantly higher than those in the blank control group(P<0.05).
