摘　　要：目的 研究蟾酥Bufonis Venenum在不同贮藏条件下化学成分的变化规律。方法 对蟾酥药材采用不同贮藏温度进行密封保存,科学规范定时采样24个月,观察其外观;HPLC法测定蟾毒灵、华蟾酥毒基和脂蟾毒配基的含量,以各成分的相对峰面积表征其变化规律。结果 随着贮藏时间的延长,-18℃、4℃环境下的蟾酥色泽未见明显变化,质地较硬;而25℃环境下的蟾酥色泽加深,质地变软。-18℃环境下蟾酥中蟾毒灵、华蟾酥毒基和脂蟾毒配基的含量基本不变;4℃、25℃环境下华蟾酥毒基含量明显下降,6个月以上蟾酥中3个指标成分含量总和低于2020年版《中国药典》规定的7.0%,主要为华蟾酥毒基转变成去乙酰华蟾酥毒基。结论 中药蟾酥应在冷冻、避光的环境下贮藏。AIM To evaluate the chemical changes of Bufonis Venenum during storage.METHODS B. Venenum stored at ambient temperature in room for 24 months was subjected to the appearance observation;the quantitative determination of bufalin, cinobufagin and resibufogenin by HPLC;the investigation of the chemical changes by analyses of the relative peak area of chemical constituents.RESULTS An extended period of time left no significant color change and hard texture to the B. Venenum stored in room at-18 ℃ and 4 ℃, but turned those stored at 25 ℃ into darker color and soft texture. B. Venenum was observed with no content changes of bufalin, cinobufagin and resibufogenin at-18 ℃;but significantly decreased content of cinobufagin, and lower than 7% total contents of bufalin, cinobufagin and resibufogenin under exposure of 6-month 4 ℃ and 25 ℃ ambient temperature because of the degradation of cinobufagin, which was inconsistent with the requirement of Chinese Pharmacopeia(2020 edition).CONCLUSION B. Venenum should be stored in a cold environment away from sunlight.
