摘  要:目的:观察壮通饮对脑缺血再灌注损伤模型大鼠脑缺血损伤的保护作用,并从时钟基因Bmal1调控昼夜节律途径初步探讨其保护作用的机制。方法:取SD大鼠60只,按6、12、18、24时四个时间点随机分为4组,每组大鼠随机分为:假手术组、模型组、壮通饮组。观察昼夜节律对脑缺血模型大鼠神经功能缺损评分,脑梗死体积,脑含水量,Bmal1蛋白表达水平的影响以及壮通饮干预后每组大鼠各指标的变化情况。结果:各组大鼠均有明显的神经功能缺损症状,其中6∶00组大鼠神经缺损症状明显重于24∶00组,24∶00组大鼠脑梗死体积明显小于6∶00组,18∶00、24∶00组大鼠脑含水量明显小于6∶00组,18∶00、24∶00组大鼠脑组织Bmal1表达量明显高于6∶00组。壮通饮干预后,24∶00组大鼠神经缺损症状较6∶00组大鼠明显减轻,18∶00、24∶00组大鼠脑梗死体积,脑含水量较6∶00,12∶00组明显下降,24∶00组大鼠脑组织Bmal1表达水平较6∶00组明显增高。结论:壮通饮可通过调控时钟基因Bmal1活性来调控昼夜节律发挥脑缺血损伤保护作用。Objective:Observe the protective effect of Zhuangtong Yin(ZTY)on cerebral ischemia injury in rats.Explore the mechanism of protective effect on the clock gene Bmal1in regulation of the circadian rhythm.Methods:Divide 60SD rats into 4groups at the four time points of 6、12、18and 24o'clock randomly.Each group of rats was randomly divided into:sham group,model group and ZTY group.Observe the effect of circadian rhythm on neurological deficit score,cerebral infarction volume,brain water content,and Bmal1protein expression level in rats with cerebral ischemia.Observe changes of various indexes of rats in each group after intervention of ZTY.Results:Rats in each group had obvious symptoms of neurological deficits.The symptoms of neurological deficits in the 6∶00group were significantly heavier than those in the 24∶00group.The cerebral infarction volume of rats in the 24∶00group was significantly smaller than that in the 6∶00group.The brain water content of rats in the 24∶00group was significantly lower than that in the 6∶00group.The expression level of Bmal1in the brain tissue of rats in the 18∶00and 24∶00groups was significantly higher than that in the 6∶00group.After the intervention of ZTY:the symptoms of nerve defects in the 24∶00group were significantly reduced compared with those in the 6∶00group.The cerebral infarction volume and brain water content of rats in the 24∶00group were significantly lower than those in the 6∶00and 12∶00groups.The expression level of Bmal1in the brain tissue of rats in the 24∶00group was significantly higher than that in the 6∶00group.Conclusion:ZTY play aprotective role against cerebral ischemia through regulate the circadian rhythm by regulating the activity of the clock gene Bmall.
