摘  要:目的:探讨宫内缺氧对幼年大鼠海马神经元Jumonji结构域蛋白(JMJD)6和突触素蛋白表达水平及对其学习记忆力的影响.方法:将40只健康SD孕鼠,随机分为缺氧1d、2d和3d组(研究组)与对照组,每组10只孕鼠.将研究组孕鼠于怀孕第14天置于低压低氧动物舱中,诱导宫内缺氧模型;Morris水迷宫实验检测幼年大鼠的空间学习记忆力;蛋白免疫印迹(WB)检测幼年大鼠海马神经元JMJD6和突触素的表达水平.结果:幼年大鼠在Morris水迷宫实验中,逃避潜伏期缺氧组较对照组明显延长(P<0.05),空间探索实验在平台象限的时间缺氧组较对照组缩短(P<0.05);WB检测显示随缺氧时间增加,幼年大鼠海马神经元JMJD6蛋白表达水平逐渐增加,突触素蛋白表达水平逐渐减少,研究组与对照组相比,差异均有统计学意义(P<0.05).结论:宫内缺氧可致SD幼年大鼠的学习记忆力降低,可能与海马神经元JMJD6表达增加致突触素表达降低有关.Objective:To investigate effects of intrauterine hypoxia on expressions of Jumonji domain-containing protein(JMJD)6 and synaptophysin protein in the hippocampus neurons of young rats and their learning and memory.Methods:Forty healthy SD pregnant rats were randomly divided into the hypoxia 1d,2d and 3d groups,control group(n=10,in each group).The intrauterine hypoxia model was established by placing the pregnant rat in a low-pressure and hypoxic experimental animal chamber.Morris water maze was used to detect the escape latency,residence time in the target quadrant and swimming trajectory of young rats.The expressions of JMJD6 and synaptophysin in the hippocampus of young rats were detected by western blot(WB).Results:In the Morris water maze test,the escape latency of young rats in the hypoxia group was significantly longer than that in the control group(P<0.05);The stay time of the space exploration experiment in the platform quadrant in the hypoxia group was shorter than that in the control group(P<O.05);WB showed that with the increase of hypoxia time,the expression of JMJD6 protein increased obviously and the expression of synaptophysin protein decreased gradually compared with the control group.(P<0.05).Conclusion:Intrauterine hypoxia can reduce young SD rats'learning and memory ability,which may be related to JMJD6 increase and synaptophysin decrease in the hippocampus.
