摘  要:2-乙基己基二苯基磷酸酯(EHDPP)是一种典型的有机磷酸酯阻燃剂,近年来在水环境和水生生物组织中有着较高的检出率.为探究低浓度EHDPP对浮游动物的发育和生殖毒性及母体传递效应,以0、0.1、1、10和100μg/L的EHDPP对甲壳类浮游生物大型溞进行三代慢性暴露,研究EHDPP暴露对大型溞生殖、发育及相关基因转录的影响.研究发现100μg/L EHDPP慢性暴露下F0大型溞体长下降,F1新生儿体长下降并出现雄性个体.继续开展F1和F2大型溞的21 d连续多代暴露试验,并设置恢复组(无EHDPP暴露)进行大型溞的21 d恢复实验.结果发现100μg/L EHDPP恢复组和连续暴露组中F1大型溞的体长均受到显著抑制,F2中均观察到雄性个体出现,说明EHDPP对大型溞生长的影响持续到了F2代.基因表达结果显示,EHDPP暴露下调了细胞循环途径相关基因的表达,上调了幼年激素酯酶基因的表达量,说明EHDPP可能通过引起细胞周期紊乱和激素水平变化进而干扰大型溞正常的生长生殖.综上所述,EHDPP长期暴露引起了大型溞的发育毒性和生殖毒性,并能对大型溞后代造成持续性影响.2-Ethylhexyl diphenyl phosphate(EHDPP), one of the typical organophosphate flame retardant(OPFRs), was frequently detected in surface water and aquatic organisms. To explore the chronic developmental toxicity, reproductive toxicity, and maternal transmission effect of EHDPP with low concentration in zooplankton, Daphnia magna were chronically exposed to 0, 0.1, 1, 10 and 100 μg/L EHDPP for three generations, respectively, and their exposure effects on reproduction, growth and the transcription of related genes were evaluated. It was found that the body length of F0 generation under chronic exposure of 100 μg/L EHDPP decreased. The gender differentiation and inhibition of newborn body length were observed in the F1 generation, and male individuals were found in the offspring. The 21-day continuous exposure experiment of Daphnia magna F1 and F2 was carried out with a 21-day recovery test(without EHDPP exposure). The results showed that the body length of F1 generation was significantly inhibited in both groups of recovery and continuous exposure. The male individuals were also observed in both groups of F2 as their newborns, indicating that inhibition of EHDPP on body length continued to the F2 generation. Gene expression results showed that EHDPP exposure down-regulated the expression of genes that related to cell cycle pathways, and increased the expression of the juvenile hormone esterase gene jhe, suggesting that EHDPP might interfere the normal growth and reproduction of Daphnia manga by disturbing cell cycle and changing hormone levels. In conclusion, long-term exposure to EHDPP could lead to developmental toxicity and reproductive toxicity of Daphnia magna, and have a continuous impact on its offspring.
