五味子水提物保护大鼠胃黏膜及抑制小鼠胃肠运动研究

摘　　要：目的考察五味子水提物保护大鼠胃黏膜及抑制小鼠胃肠运动的作用。方法大鼠60只,随机分为6组,分别为正常对照组,模型对照组,五味子水提物高、中、低剂量组,奥美拉唑阳性对照组,每组10只。五味子水提物高、中、低剂量组按1.4 g/kg、0.7 g/kg、0.35 g/kg剂量分别给予五味子水提物灌胃,正常对照组、模型对照组大鼠给予等体积蒸馏水灌胃,奥美拉唑阳性对照组按20 mg/kg剂量灌胃。给药10 d后,以乙醇诱发大鼠胃溃疡模型,然后检测五味子水提物对大鼠溃疡的抑制率及大鼠胃壁细胞病理改变的影响。同时采用正常小鼠和新斯的明所致胃肠功能亢进小鼠模型,考察五味子水提物对胃肠运动功能的影响。结果五味子水提物能显著减少乙醇所致大鼠胃溃疡的面积,1.4 g/kg、0.7 g/kg、0.35 g/kg水提取物的胃溃疡抑制率分别为(88.7±9.3)%、(71.8±13.3)%、(28.8±11.5)%。组织病理学检查可见,给予五味子水提物后,大鼠胃壁病变严重程度减轻,炎性细胞浸润程度减轻。五味子水提物可抑制小鼠胃排空及肠推进,尤其对新斯的明所致胃肠功能亢进小鼠的抑制作用更为明显,与模型组比较,五味子水提物高、中剂量组以及颠茄阳性对照组小鼠的小肠推进率明显降低(P<0.05),胃残留率显著增加(P<0.05)。结论五味子水提物不仅可保护胃黏膜,抑制溃疡发生,同时可抑制胃肠运动,对于伴有腹泻、胃肠痉挛的消化性溃疡可能具有良好疗效。Objective To investigate the protective effects of Schisandra chinensis water extract on gastric mucosal injury in rats and gastrointestinal motility in mice.Methods Sixty rats were randomly divided into 6 groups:normal control group,model control group,Schisandra chinensis extract high-dose,medium-dose and low-dose groups,and omeprazole positive control group,with 10 rats in each group.The rats in the high-dose,medium-dose and low-dose groups were given Schisandra chinensis water extract by gavage at the doses of 1.4 g/kg,0.7 g/kg and 0.35 g/kg,respectively.The rats in the normal control group and the model control group were given the same volume of distilled water by gavage.The rats in the omeprazole positive control group were given gavage at the dose of 20 mg/kg.After 10 days of administration,ethanol was used to induce gastric ulcer model in rats,and then the inhibitory rate of Schisandra chinensis extract on ulcer and the pathological changes of gastric parietic cells in rats were detected.The effects of water extract of Schisandra chinensis on gastrointestinal motility were investigated in normal mice and neodemine induced gastrointestinal hyperfunction mice.Results Schisandra water extract could significantly reduce the area of gastric ulcer induced by ethanol in rats,and the inhibition rates at the doses of 1.4,0.7 and 0.35 g/kg were(88.7±9.3)%,(71.8±13.3)%and(28.8±11.5)%respectively.Histopathological examination showed that after Schisandra chinensis water extract was given,the severity of gastric wall lesions and inflammatory cell infiltration in gastric ulcer rats were reduced.Schisandra chinensis water extract can inhibit gastric emptying and intestinal propulsion in mice;especially in mice with gastrointestinal hyperfunction induced by neostigmine,which compared with the model group,the small intestinal propulsion rate of mice in the high and medium dose groups of Schisandra water extract decreased significantly(P<0.05),and the gastric residue rate increased significantly(P<0.05).Conclusion Schi
