摘 要:哺乳动物雷帕霉素靶蛋白(mTOR)属于进化上高度保守的丝氨酸/苏氨酸蛋白激酶,可以对细胞生长、代谢、存活和免疫反应等几个基本生物学功能进行关键调控。在以往的临床试验中,mTOR信号抑制被认为是胃肠道间质瘤有希望的靶向治疗策略,但目前利用该靶点治疗胃肠道间质瘤的研究大多只是单一作用该通路或靶点,其疗效并不令人满意。由于胃肠道间质瘤等肿瘤性疾病发病机制涉及多条通路,因此,联合mTOR等信号通路的靶向治疗方案可能是未来治疗晚期胃肠道间质瘤主要方法。Mammalian target of rapamycin(mTOR)is an evolutionarily highly conserved serine/threonine protein kinase that regulates several basic biological functions,including cell growth,metabolism,survival,and immune response.In previous clinical trials,mTOR signaling inhibition was considered as a promising targeted therapy strategy for gastrointestinal stromal tumors.However,most of the current studies using this target for the treatment of gastrointestinal stromal tumors only targeted this pathway or target,and its efficacy was not satisfactory.Since the pathogenesis of tumor diseases such as gastrointestinal stromal tumor involves multiple pathways,targeted therapy combined with mTOR or oher signal pathways may be the main method for the treatment of advanced gastrointestinal stromal tumor in the future.
