摘 要:对破壁灵芝孢子粉破壁率的检测方法进行优化,以降低试验误差。使用血球计数板对灵芝孢子粉和破壁灵芝孢子粉进行计数,考虑到灵芝孢子粉破壁率的分析方法中出现的孢子悬液分布不均匀问题,故对超声振荡的时间和蔗糖粉末粗细程度试验条件进行优化,并对优化前后的结果进行比较。结果表明,方法优化前,破壁灵芝孢子粉破壁率测量为98%,相对标准偏差(S)为10.25%,经试验条件优化后,其破壁率测量的结果提高到99%,相对标准偏差(S)下降至1.79%,前后差异明显,效果满意。通过分析灵芝孢子粉破壁率测量误差的主要来源,并采取针对性措施,可以有效降低灵芝孢子粉计数结果的相对标准偏差,提升破壁率测量的准确度。To optimize the detection method of wall breaking rate of Ganoderma lucidum spore powder and to reduce experimental error,Ganoderma lucidum spore powder and wall broken Ganoderma lucidum spore powder were counted by blood cell counting plate.Considering the uneven distribution of spore suspension in the analysis method of wall breaking rate of Ganoderma lucidum spore powder,the experimental condition of ultrasonic oscillation time and the thickness of sucrose powder was optimized,and the results before and after optimization were compared.Before the optimization of the method,the measurement results of the wall breaking rate of Ganoderma lucidum spore powder were 98%,and the relative standard deviation (S) was 10.25%.After the optimization of experimental condition,the measurement results of the wall breaking rate improved to 99%,and the relative standard deviation (S) reduced to 1.79%.By analyzing the main sources of measuring error of Ganoderma lucidum spore powder breaking rate and taking targeted measures,the relative standard deviation of Ganoderma lucidum spore powder counting results could be effectively reduced and the accuracy of measuring wall breaking rate could be improved.
