摘 要:目的 观察共济失调毛细血管扩张突变(ataxia telangiectasia-mutated,ATM)基因rs11212617多态性对2型糖尿病二甲双胍疗效的影响。方法 选取2015年10月—2017年12月泰兴市人民医院新诊断的60例2型糖尿病患者给予二甲双胍治疗,检测治疗前及治疗后血糖、糖化血红蛋白及生化指标,并进行ATM基因检测。结果 ATM基因rs-11212617多态性有:AA型、AC型、CC型。治疗前各基因型患者空腹血糖(FPG)、餐后2 h血糖(2 hPG)、糖化血红蛋白(HbA1c)水平比较,差异无统计学意义(P>0.05)。治疗12周后各组患者FPG、2 hPG、HbA1c水平下降,AA组FPG、2 hPG、HbA1c下降幅度大于AC组及CC组,差异有统计学意义(P<0.05),且各组治疗前后差值比较,差异有统计学意义(P<0.05)。结论 ATM基因rs11212617多态性可能与二甲双胍降糖疗效有关,AA基因型患者对二甲双胍的反应优于AC、CC基因型。Objective To observe the effect of ataxia telangiectasia-mutated(ATM)gene rs11212617 polymorphism on the efficacy of metformin in type 2 diabetes.Methods Sixty patients with type 2 diabetes newly diagnosed in Taixing People´s Hospital from October 2015 to December 2017 were selected and given metformin treatment.Blood glucose,glycosylated hemoglobin and biochemical indexes were detected before and after treatment,and ATM gene detection was performed.Results The rs-11212617 polymorphisms of ATM gene were:AA type,AC type and CC type.Before treatment,there was no statistically significant difference in the levels of fasting blood glucose(FPG),2 h postprandial blood glucose(2 hPG)and glycosylated hemoglobin(HbA1c)between patients with different genotypes(P>0.05).After 12 weeks of treatment,the levels of FPG,2 hPG,and HbA1c in each group decreased,and the decrease in FPG,2 hPG,and HbA1c in the AA group was greater than that in the AC and CC groups,and the difference was statistically significant(P<0.05).And the difference of each group before and after treatment was compared,and the difference was statistically significant(P<0.05).Conclusion The rs11212617 polymorphism of ATM gene may be related to the hypoglycemic effect of metformin,and the response of AA genotype to metformin may be better than that of AC and CC genotypes.
