摘  要:目的 卵巢癌(OC)为全世界女性发病率最高的癌症之一。环状RNA(circRNA)稳定性高,保守性强,可作为微小RNA(miRNA)分子的“海绵”,控制miRNA下游的基因表达与突变。本研究旨在探索circ-102958在卵巢癌中的表达和对miR-1205及其下游基因SH2D3A的调控关系,为卵巢癌早期诊断及治疗的临床转化提供理论基础,提高卵巢癌患者生存率。方法 CCK-8实验分组观察circ-102958/miR-1205/SH2D3A轴对卵巢癌细胞增殖能力的影响,Transwell实验分组观察circ-102958/miR-1205/SH2D3A轴对卵巢癌细胞迁移和侵袭的影响。结果 circ-102958表达水平和卵巢癌分期显著相关。circ-102958能够通过调节miR-1205促进SH2D3A的表达,促进卵巢癌细胞的增殖、迁移和侵袭。结论 circ-102958与患者恶性表型和预后有关,是潜在的癌基因。circ-102958能够通过调控miR-1205促进SH2D3A的表达,从而促进卵巢癌的进展。Objective Ovarian cancer(OC) is one of the highest incidence of cancers in women in the world. Circular RNA(circRNA) is highly stable and conserved, and can be used as the “sponge” of miRNAs molecules to control the gene expression and mutations downstream of miRNA. This study aims to explore the expression of circ-102958 in ovarian cancer and its regulatory relationship with miR-1205 and its downstream gene SH2D3A, so as to provide theoretical basis for the clinical transformation of the early diagnosis and treatment of ovarian cancer, and improve the survival rate of ovarian cancer patients. Methods The effects of the circ-102958/miR-1205/SH2D3A axis on proliferation ability of ovarian cancer cells were observed in CCK-8 group, and the effects of circ-102958/miR-1205/SH2D3A axis on migration and invasion of ovarian cancer cells were observed in Transwell group. Results The expression level of circ-102958 was significantly correlated with ovarian cancer stage. circ-102958 can promote the expression of SH2D3A by regulating miR-1205, and promote the proliferation,migration and invasion of ovarian cancer cells. Conclusion circ-102958 is a potential oncogene associated with clinical malignant phenotype and prognosis. circ-102958 promotes the expression of SH2D3A by regulating miR-1205, thereby promoting the progression of ovarian cancer.
