摘 要:生鲜食品的检验技术关系到我国的食品安全问题,现阶段还需要加强并完善相应的检测技术。微生物酶技术中的酶联免疫法(Enzyme-Linked ImmunoSorbent Assay,ELISA)可以用于食品中拟除虫菊酯类农药的残留检测。试验利用间接竞争ELISA技术对拟除虫菊酯类农药及其代谢产物的检测,并对该方法进行了优化。优化后的拟除虫菊酯类检测条件:离子强度10 mmol/L,pH值7.4,10%甲醇,标准曲线的线性范围2.65~28.55 ng/mL,灵敏度为8.69~344.97 ng/mL;优化后的代谢产物检测条件:包被浓度125 ng/mL,抗体稀释倍数1︰2000,pH值6.6,吐温浓度0.005%,离子浓度40 mmol/L,S-HPR稀释倍数1︰4000,标准曲线的线性范围为0.37~7.40 ng/mL,检测限为0.15 ng/mL。结果表明,试验建立的间接竞争ELISA可以有效地进行多种拟除虫菊酯类农药及其代谢产物的残留检测,且准确性和灵敏度高。The inspection technology of fresh food is related to the food safety problem in our country.At present,we need to strengthen the establishment and improvement of testing technology.Enzyme-linked immunosorbent assay(ELISA)in microbial Enzyme technology can be used to detect pyrethroid pesticide residues in food.In this experiment,the indirect competitive ELISA technology was used to detect pyrethroid pesticides and their metabolites,and the method was optimized.The optimized conditions for pyrethroid detection were as follows:ionic strength 10 mmol/L,pH 7.4,10%methanol,2.65~28.55 ng/mL of standard curve linear range and 8.69~344.97 ng/mL of sensitivity.Optimized metabolite detection conditions were as follows:coating concentration 125 ng/mL,antibody dilution 1︰2000,pH 6.6,tween concentration 0.005%,ion concentration 40 mmol/L,S-HPR dilution 1︰4000,0.37~7.40 ng/mL of standard curve linear range and 0.15 ng/mL of detection limit.The results showed that the established indirect competitive ELISA could effectively detect the residues of pyrethroid pesticides and their metabolites with high accuracy and sensitivity.
