摘  要:本试验旨在探究反式茴香脑(AN)对肉鸡体内外抗氧化能力的影响。体外试验:选用强抗氧化剂二丁基羟基甲苯(BHT)作为阳性对照,采用1,1-二苯基-2-苦基肼(DPPH)自由基清除法、2,2′-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基清除法、羟基自由基清除法和铁离子还原能力(FRAP)法系统评价AN的抗氧化能力,AN纯品的抗氧化能力表示为半抑制浓度(IC50)。体内试验:选取1日龄爱拔益加(AA)肉仔鸡256只,随机分为4组,每组8个重复,每个重复8只鸡。4个组在基础饲粮中分别添加0、200、400和600 mg/kg的AN(分别为AN 0、AN 200、AN 400和AN 600组)。试验期共42 d,分为生长前期(1~21日龄)和生长后期(22~42日龄)。结果表明:1)AN清除DPPH、ABTS和羟自由基的IC50分别为16.26、6.98和7.82 mg/mL,对三价铁离子的还原能力较弱。2)与AN 0组相比,AN 400组22~42日龄平均日采食量(ADFI)显著提高(P<0.05),AN 200组1~42日龄ADFI显著提高(P<0.05);AN对平均日增重(ADG)和料重比(F/G)无显著影响(P>0.05)。21日龄抗氧化能力,与AN 0组相比,AN 200、AN 400和AN 600组血清总超氧化物歧化酶(T-SOD)活性均显著升高(P<0.05),AN400和AN 600组血清丙二醛(MDA)含量显著降低(P <0.05);AN 600组肝脏过氧化氢酶(CAT)活性显著升高(P<0.05),AN 200和AN 400组肝脏T-SOD活性显著升高(P<0.05);AN200、AN 400和AN 600组肝脏CAT的基因相对表达量显著提高(P<0.05),AN 400和AN 600组肝脏超氧化物歧化酶1(SOD1)的基因相对表达量显著升高(P<0.05),AN 600组肝脏血红素加氧酶-1(HO-1)的基因相对表达量显著升高(P<0.05)。42日龄抗氧化能力,与AN 0组相比,AN 400和AN 600组血清总抗氧化能力(T-AOC)显著升高(P<0.05);AN 200和AN 400组肝脏CAT活性显著升高(P<0.05),AN 200组胸肌谷胱甘肽过氧化物酶(GSH-Px)活性、T-AOC显著提高(P<0.05);AN 200组胸肌谷胱甘肽过氧化酶1(GPx1)的基因相对表达量显著提高(P<0.05)。综上所述,AN有一定的�This study was conducted to explore the antioxidant capacity of trans-anethole(AN)of AA broilers in vivo and in vitro.In vitro test,the strong antioxidant butylated hydroxytoluene(BHT)was used as a positive control,and the antioxidant activity of AN was evaluated by 1,1-diphenyl-2-picrylhydrazyl(DPPH),diammonium 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate)(ABTS),hydroxyl radical scavenging methods and ferric ion reducing antioxidant power(FRAP)method,the antioxidant capacity of AN was expressed as half maximal inhibitory concentration(IC50).In vivo test,a total of 256 healthy 1-day-old broilers were randomly divided into four groups,with eight replicates in each group and eight chickens in each replicate.The addition levels of AN in each group were 0,200,400 and 600 mg/kg(AN 0,AN 200,AN 400 and AN 600groups).The experimental period was 42 days,which was divided into two stages:the early stage(1 to 21days of age)and the late stage(22 to 42 days of age).The results showed as follows:1)the IC50values of AN for DPPH,ABTS and hydroxyl radicals were 16.26,6.98 and 7.82 mg/mL.The reduction ability for Fe3+was weak.2)Compared with AN 0 group,the average daily feed intake(ADFI)of AA broilers aged 42 days in AN 400 group significantly increased(P0.05).Effects of AN on the antioxidant capacity of AA broilers aged 21 days:compared with AN 0 group,the serum total superoxide dismutase(T-SOD)activity in AN 200,AN 400 and AN 600 groups significantly increased(P <0.05),and the serum malondialdehyde(MDA)content in AN 400 and AN 600 groups significantly decreased(P<0.05),the liver catalase(CAT)activity in AN 600 group significantly increased(P<0.05),and the TSOD activity in AN 200 and AN 400 groups significantly increased(P<0.05),the relative expression level of catalase(CAT)gene in liver in AN 200,AN 400 and AN 600 groups significantly increased(P<0.05),and the relative expression level of superoxide dismutase 1(SOD1)gene in AN 400 and AN 600 groups significantly increased(P<0.05),the relative expression level of heme oxyg
