摘  要:目的探讨止痛消结散治疗癌性疼痛的作用机制。方法从小鼠体内成功分离出背根神经节(Dorsal root ganglion,DRG)神经元细胞,在此基础上构建肿瘤细胞与DRG神经元共培养模型及药物干预模型,选取10倍稀释的止痛消结散浸提液进行ELISA、RT-PCR和Western Blot实验,利用ELISA技术检测模型中IL-6的表达水平,利用Western Blot检测模型中DRG神经元的TRPV1蛋白表达水平,利用RT-PCR检测模型中DRG细胞的IL-6R、Trpv1 mRNA表达水平。结果止痛消结散药物干预模型组中IL-6、IL-6R、Trpv1 mRNA表达水平显著下降,目的蛋白TRPV1表达水平显著降低。结论止痛消结散能有效缓解癌性疼痛,其作用机制可能与下调DRG神经元中TRPV1表达水平有关。Objective To explore the mechanism of Zhitong Xiaojie Powder in the treatment of cancer pains.Methods The dorsal root ganglion(DRG)neurons were successfully isolated from mice,based on which a co-culture model of tumor cells and DRG neurons and a drug intervention model were constructed.The 10 times diluted extract of Zhitong Xiaojie Powder was selected for ELISA,RT-PCR and Western blot experiments.The expression levels of IL-6 in the models were detected by ELISA,the TRPV1 protein expression levels in DRG neurons were detected by Western blot,and the IL-6R,Trpv1 mRNA expression levels in DRG cells in the models were detected by RT-PCR.Results The expression levels of IL-6,IL-6R,Trpv1 mRNA and the expression level of target protein TRPV1 were significantly decreased in the model group treated with Zhitong Xiaojie Powder.Conclusion Zhitong Xiaojie Powder can effectively relieve cancer pains,which may be related to the down-regulation of TRPV1 expression in DRG neurons.
