摘 要:为鉴定糯玉米品种的真实性,探究糯玉米品种间的亲缘关系。本研究以国内市场上推广的39份糯玉米品种为试材,利用基于靶向测序基因型检测(genotyping by target sequencing,GBTS)技术进行SNP(single nucleotide polymorphisms,SNP)标记检测,并开展DNA指纹图谱构建和NJ聚类分析。结果表明,在39份品种中获得6397个SNP基因分型数据,根据基因型完整度和次要等位基因频率(minor allele frequency,MAF)筛选了4767个SNP位点,平均杂合率为33.94%;缺失率平均值为0.54%;MAF值分布在0.054~0.500之间,平均值为0.260;多态性信息含量(polymorphism information content,PIC)分布在0~0.375之间,PIC平均值为0.28,为中度多态性位点;39份糯玉米品种的SNP杂合率为20.62%~29.94%,平均值为26.79%,具有较高的杂合度;通过筛选PIC值大于0.37的25个SNP位点,可完全区分39个品种,鉴定率100%;基于4767个SNP的聚类分析结果表明,39个品种被划分为5大类群。本研究结果可为糯玉米品种鉴定、自交系选育提供理论和技术支持。In order to identify the authenticity of waxy corn varieties and explore the genetic relationship between waxy corn varieties.In this study,39 waxy corn varieties promoted in the domestic market were used as experimental materials,single nucleotide polymorphisms(SNP)markers were detected using the technique of genotyping by target sequencing(GBTS),and DNA fingerprinting and NJ clustering analysis were carried out.The results showed that 6397 SNPs genotyping data were obtained from 39 varieties,and 4767 SNPs were screened according to the genotype integrity and minor allele frequency(MAF).The average heterozygosity was 33.94%.The average deletion value was 0.54%.The MAF values were distributed between 0.054~0.500,with an average of 0.260.The polymorphism information content(PIC)values were distributed in the range of 0~0.375,with an average PIC of 0.28,which were moderate polymorphism loci.The SNP heterozygosity of 39 waxy maize varieties were 20.62%~29.94%,with an average of 26.79%,with high heterozygosity.By screening 25 SNP loci with PIC val ues greater than 0.37,39 varieties could be completely distinguished,and the identification rate was 100%.Clustering analysis based on 4767 SNPs showed that 39 varieties were divided into five groups.The results of this study can provide theoretical and technical support for waxy corn varieties identification and inbred lines breeding.
