摘  要:目的:探讨B细胞淋巴瘤-2(Bcl-2)基因与信号转导子和转录激活子3(STAT3)在前列腺癌组织中的表达情况、相关性及其临床意义。方法:选取21例前列腺增生和27例前列腺癌患者的石蜡标本及收集其临床资料,分为前列腺增生组和前列腺癌组,应用免疫组化、免疫荧光法检测在前列腺增生、前列腺癌组织中STAT3与Bcl-2的表达。结果:STAT3、Bcl-2蛋白在前列腺癌组织的表达率均高于前列腺增生组织(96.3%vs.33.3%、96.3%vs.28.6%,P<0.05);两者表达与病理分型、临床分期、Gleason评分及血清PSA具有相关性(P<0.05),与年龄不具有相关性(P>0.05)。低分化、临床分期为Ⅲ+Ⅳ期前列腺癌患者的Bcl-2和STAT3阳性表达率均高于高分化、临床分期为Ⅱ期的前列腺癌患者(P<0.05)。Spearman等级相关分析显示前列腺癌组织中STAT3与Bcl-2表达呈正相关(r=0.685,P<0.05)。结论:前列腺癌患者的Bcl-2和STAT3阳性表达率均较高,与前列腺癌的分化程度和分期相关,二者可能存在一定的相互协同作用,Bcl-2与STAT3可能通过某种方式共同调控前列腺癌的发生发展。Objective:To investigate the expression, correlation, and clinical significance of B-cell lymphoma-2 gene(Bcl-2) and signal transducer and activator of transcription(STAT3) in prostate cancer.Methods:Paraffin specimens and clinical data of 21 patients with benign prostatic hyperplasia and 27 patients with prostate cancer were selected and divided into prostate hyperplasia group and prostate cancer group.The expression of STAT3 and Bcl-2 in the tissues of benign prostatic hyperplasia(BPH) and prostate cancer(PCa) was detected by immunohistochemistry and immunofluorescence.Results:STAT3 and Bcl-2 proteins were highly expressed in prostate cancer tissues than in prostate hyperplasia tissues(96.3% vs.33.3%,96.3% vs.28.6%,P<0.05).,The expressions were correlated with pathological staging, clinical stage, Gleason score and serum PSA(P<0.05),there was no correlation with age(P>0.05).Both Bcl-2 and STAT3 positive expression rates in prostate cancer patients with low differentiation and clinical stage III + IV were higher than those in prostate cancer patients with high differentiation and clinical stage II(P<0.05).Spearman rank correlation analysis showed STAT3 and Bcl-2 were positively correlated in prostate cancer tissues(r= 0.685,P<0.05).Conclusion:The positive expression of both Bcl-2 and STAT3 in prostate cancer patients is high and correlates with the degree of differentiation and stage of prostate cancer, and there may be some mutual synergistic effects between the two, and Bcl-2 and STAT3 may co-regulate the development of prostate cancer in some way.
