摘  要:[目的]探讨溃疡性结肠炎患者肠黏膜组织中miR-24的表达与肠道菌群和炎症因子的关系。[方法]选取93例溃疡性肠黏膜患者,并根据病情分为活动期溃疡性结肠炎组(活动期组,48例)和缓解期溃疡性结肠炎组(缓解期组,45例),另选同期因结肠息肉切除肠黏膜组织的患者50例作为对照组,通过RT-qPCR检测各组肠黏膜组织中miR-24的表达水平,分析miR-24与溃疡性结肠炎患者临床病理参数的关系。ELISA试剂盒检测各组人群血清炎症因子(IL-6、TNF-α)含量,检测各组粪便中肠道菌群(肠杆菌、肠球菌、乳酸杆菌、双歧杆菌)数量。Pearson相关分析miR-24与炎症因子和肠道微生物的关系。[结果]对照组、缓解期组和活动期组患者肠黏膜组织miR-24表达水平分别为1.79±0.46、2.56±0.52、5.74±0.79,各组间比较差异有统计学意义(F=579.551,P<0.05);且miR-24的表达水平与疾病程度有关(F=4.520,P<0.05)。ELISA结果显示:与对照组比较,缓解期组和活动期组患者血清IL-6、TNF-α含量显著升高(F=824.899、F=441.239,P<0.001);粪便中肠杆菌、肠球菌数量显著增加,双歧杆菌、乳酸杆菌数量显著减少(F=670.031,F=490.763,F=320.209,F=1032.835,均P<0.001)。Pearson相关分析结果显示:miR-24的表达与活动期患者血清IL-6、TNF-α、粪便肠杆菌、肠球菌呈正相关(P<0.05),与粪便乳酸杆菌、双歧杆菌呈负相关(P<0.05)。[结论]miR-24高表达与溃疡性结肠炎患者炎症因子和肠道菌群失调有关。[Objective]To explore the relationship between the expression of miR-24 in the intestinal mucosa of patients with ulcerative colitis and the intestinal flora and inflammatory factors.[Methods]Ninety-three patients with ulcerative intestinal mucosa were selected and divided into active ulcerative colitis group(active phase group: 48 cases)and remission stage ulcerative colitis group(remission stage group: 45 cases),and 50 patients with intestinal mucosal tissues removed due to colon polyps during the same period were selected as the control group.The expression of miR-24 in intestinal mucosa was detected by RT-qPCR,and the relationship between miR-24 and clinicopathological parameters of patients with ulcerative colitiswere analyzed.The serum levels of IL-6 and TNF-α were detected by ELISA kit.The number of enterobacter, Enterococcus, Lactobacillus and Bifidobacterium in feces of the three groups were detected.Pearson Correlation analysis was used to analyze the relationship between miR-24 and inflammatory factors and intestinal microflora.[Results]The expression levels of miR-24 in intestinal mucosa of patients in control group, remission group and active stage group were 1.79±0.46,2.56±0.52 and 5.74±0.79,respectively, with significant difference among the three groups(F=579.551,P<0.05);and the expression level of miR-24 was related to the degree of disease(F=4.520,P<0.05).The results of ELISA showed that compared with the control group, the levels of serum IL-6 and TNF-α in remission group and active stage group were significantly increased(F=824.899,F=441.239,P<0.001);the number of Enterobacteriaceae and Enterococcus in feces was significantly increased, while the number of Bifidobacterium and Lactobacillus in feces was significantly decreased(F=670.031,F=490.763,F=320.209,F=1032.835,allP<0.001).The results of Pearson correlation analysis showed that the expression of miR-24 was positively correlated with serum IL-6,TNF-α,Enterobacter faecalis, and Enterococcus(P<0.05),while negatively correlated with Lac
