摘 要:目的探讨环状RNA circ_0000591(has circ 0000591)在乙型肝炎病毒(HBV)感染相关肝癌患者血清中的表达及其沉默对HepG2.2.15细胞凋亡的影响。方法收集2019年5月-2021年5月新疆医科大学附属肿瘤医院收治的83例HBV相关肝癌患者作为肝癌组;同期收集83例HBV相关肝硬化患者为肝硬化组。通过患者电子病历系统收集患者性别、年龄、体质量指数(BMI)和基础疾病。采用生化自动分析仪检测血清丙氨酸氨基转移酶(ALT);化学发光法测定血清甲胎蛋白(AFP);双抗体夹心法检测乙型肝炎表面抗原(HBsAg);荧光定量聚合酶链式反应(RT-qPCR)检测血清HBV-DNA水平。采用Pearson相关分析血清has circ 0000591与ALT、HBsAg、HBV-DNA及AFP水平相关性。构建沉默has circ 0000591 HepG2.2.15细胞模型,设置siRNA组、NC组和control组。AnnexinV-FITC/PI双染法检测细胞凋亡。结果肝癌组has circ 0000591、ALT、AFP水平高于肝硬化组,HbsAg、HBV-DNA水平低于肝硬化组(P<0.05);两组患者血清has circ 000059水平与血清HBsAg和HBV-DNA呈负相关(P<0.05);肝癌组患者血清has circ 000059水平与血清AFP呈正相关(P<0.05);两组患者血清has circ 000059水平与血清ALT无相关关系;siRNA组细胞凋亡率高于control组和NC组(P<0.05)。结论血清has circ 0000591在HBV感染相关性肝癌中高表达;沉默has_circ 0000591可诱导肝癌HepG2.2.15细胞凋亡,可能作为临床潜在评估肝脏损伤的生物标志物。OBJECTIVE To investigate the expression of cirRNA cirC_0000591(has circ 0000591)in serum of patients with liver cancer associated with hepatitis B virus(HBV)infection and the effect of silencing on apoptosis of HepG2.2.15 cells.METHODS A total of 83 HBV-related liver cancer patients admitted to Affiliated Cancer Hospital of Xinjiang Medical University from May 2019 to May 2021 were enrolled as liver cancer group.Eighty-three patients with HBV-related cirrhosis were included in the cirrhosis group.Gender,age,body mass index(BMI)and underlying diseases were collected through electronic medical records.Serum alanine aminotransferase(ALT)was detected by biochemical automatic analyzer.Serum ALPHA-fetoprotein(AFP)was determined by chemiluminescence.Hepatitis B surface antigen(HBsAg)was detected by double antibody sandwich method.Serum HBV-DNA levels were detected by fluorescence quantitative PCR(RT-QPCR).The correlation between serum hascirc0000591 and ALT,HBsAg,hbv-dna and AFP was analyzed by Pearson correlation analysis.Silenced HascirC0000591 HepG2.2.15 cell model was constructed,and siRNA group,NC group and control group were set.Apoptosis was detected by Annexinv-FITC/PI double staining.RESULTS The levels of hascirc0000591,ALT and AFP in HCC group were higher than those in cirrhosis group,while the levels of HbsAg and hbv-dna were lower than those in cirrhosis group(P<0.05).Serum HASCIRC000059 level was negatively correlated with serum HBsAg and HBV-DNA(P<0.05).There was a positive correlation between serum AFP and hascirc000059 in HCC group(P<0.05).There was no correlation between serum hascirc000059 level and serum ALT in 2 groups.The apoptosis rate of siRNA group was higher than that of control group and NC group(P<0.05).CONCLUSION serum hascirc0000591 is highly expressed in hbv-associated liver cancer.Silencing HAScirC0000591 can induce apoptosis of hepatocellular carcinoma HepG2.2.15 cells,which may be a potential biomarker for clinical evaluation of liver injury.
