摘  要:为了明确猪TGF-β基因家族的功能和进化关系,运用HMMER并结合BLAST对猪的TGF-β基因家族成员进行鉴定,运用多种生物信息学方法对序列进行分析,运用转录组数据分析TGF-β基因家族的组织表达规律,并对TGF-β基因家族进行功能富集分析。结果表明:猪TGF-β基因家族共有36个成员,编码蛋白质的氨基酸数量在153~719,相对分子质量在16.44~79.41 kDa,等电点在4.93~11.78,均为亲水性蛋白。二级结构以无规则卷曲为主,其次是α-螺旋。系统进化树分析表明TGF-β基因家族分为三大亚类,猪TGF-β基因家族成员与人和小鼠的TGF-β基因家族成员分别聚集,说明其进化关系较近。基因结构和结构域中同一类群的不同TGF-β基因家族成员具有较高的相似度。转录组数据显示大多数TGF-β基因家族成员在成年公猪和母猪27种不同组织和外周血单核细胞中均有表达,其中GDF1和TGFB2基因在成年猪所有组织中高表达。功能富集分析发现TGF-β基因家族参与众多的生物学过程和病理过程。研究结果为深入解析猪TGF-β基因家族的生物学功能奠定基础。In order to clarify the function and evolutionary relationship of the transforming growth factorβ(TGF-β)gene family of pigs(Susscrofa),the TGF-βgene family members were identified by combination of HMMER and BLAST,and the member sequences were analyzed using a variety of bioinformatics methods.The transcriptome data wereused to analyze the expression patterns of the genes in the different tissues of pigs.The TGF-βgene families were analyzed for functional enrichment.The results showed thata total of 36 members of the TGF-βgene family were identified in the pigs.The number of amino acids encodingproteinswas 153-719,their molecular weight was 16.44-79.41 kDa,and their isoelectric point was 4.93-11.78.All the family members werehydrophilic proteins.The secondary structure was mainly irregularly coiled,followed byα-helix.The phylogenetic tree analysis showedthat the TGF-βgene familieswere divided into 3 groups.The members of the pig TGF-βgene family and the members of the human and mouse TGF-βgene familiesgathered separately,indicating that their evolutionary relationship wasratherclose.The different TGF-βgene family members of the same group in the gene structure and domain hadhigh similarity.The transcriptome data showed that most TGF-βgene family members were expressed in 27 different tissues and the peripheral blood mononuclear cells of adult boars and sows,among which GDF1 and TGFB2 werehighly expressed in all the tissues of the adult pigs.The functional enrichment analysis revealed that the TGF-βgene family members wereinvolved in many biological and pathological processes.The results of this study provided theoretical guidance for further research on the functions of the TGF-βgene family.
