摘 要:目的建立一种能模拟人类慢性阻塞性肺病的动物模型。方法通过多次气道内滴入脂多糖(lipopolysaccharide,LPS)联合被动烟熏的方法来制备慢性阻塞性肺病大鼠模型,多角度对其进行评价。结果模型组大鼠体重增长缓慢(P﹤0.01);气道阻力明显增大(P﹤0.01);气道壁及肺组织有大量炎细胞浸润,各级支气管上皮细胞明显损伤(P﹤0.01),肺泡过度扩大融合;杯状细胞增生,气道内分泌物增多(P﹤0.01);肺组织中NE及MUC5AC蛋白表达显著增加(P<0.05)。结论通过多次气道内滴入脂多糖和被动烟熏的方式能够建立较符合人类慢性阻塞性肺病病变的大鼠模型,为进一步临床研究提供了参考依据。Objective To establish an animal model that can simulate human chronic obstructive pulmonary disease.Methods Rat model of chronic obstructive pulmonary disease(COPD)was prepared by repeated infusion of lipopolysaccharide(lipopolysaccharide,LPS)and passive fumigation.Results The weight of rats in the model group increased slowly(P﹤0.01),airway resistance increased significantly(P﹤0.01),airway wall and lung tissue were infiltrated with inflammatory cells,bronchial epithelial cells were damaged significantly(P﹤0.01),goblet cells and endocrine fluid in the airway increased(P﹤0.01),The expression of NE and MUC5 AC protein in lung tissue increased significantly(P<0.05).Conclusion Lipopolysaccharide and passive fumigation can be used to establish a rat model in accordance with human chronic obstructive pulmonary disease,which provides a reference for further clinical research.
