摘 要:目的:回顾性研究肺腺癌移植瘤裸鼠模型组织中缺氧诱导因子-1α(HIF-1α)、血清蛋白[Albumin、免疫球蛋白M(IgM)]和淋巴管内皮细胞透明质酸受体1(LYVE-1)蛋白表达及意义。方法:在裸鼠背部皮下注射肺腺癌A549细胞,建立移植瘤模型。利用活体冷冻技术(IVCT)制备标本,采用HE染色及免疫组织化学(IHC)染色分析HIF-1α、Albumin、IgM和LYVE-1蛋白的表达。结果:在A549移植瘤细胞中HIF-1α蛋白表达呈阴性。肺腺癌侵袭组织中Albumin和IgM蛋白呈阳性的功能血管密度高于非侵袭组织[(14.80±1.03)个/高倍视野(HPF)与(3.50±0.53)个/HPF,P<0.01]。肺腺癌侵袭组织中LYVE-1蛋白呈阳性的淋巴管密度高于非侵袭组织[(25.90±4.43)个/HPF与(3.60±0.52)个/HPF,P<0.01]。结论:HIF-1α蛋白表达与肺腺癌侵袭和功能性血管、淋巴管密度无关,但是功能性血管和淋巴管密度与肺腺癌的侵袭、发展密切相关。Objective:To retrospectively study the expression and significance of hypoxia inducible factor-1α(HIF-1α),serum protein[Albumin,immunoglobulin M(IgM)]and lymphatic vessel endothelial hyaluronic acid receptor-1(LYVE-1)protein in xenograft tumoRof lung adenocarcinoma in nude mice.Methods:Lung adenocarcinoma A549 cells were injected subcutaneously into the back of nude mice to establish a lung adenocarcinoma xenograft tumoRmodel.Samples were prepared by in vivo cryotechnique(IVCT),and HIF-1α,Albumin,IgM and LYVE-1 protein expression were analyzed by HE staining and immunohistochemistry(IHC)staining.Results:HIF-1αprotein was negatively expressed in A549 xenograft lung adenocarcinoma cells.The density of functional blood vessels with positive Albumin and IgM proteins in invasive tissues of lung adenocarcinoma were higheRthan those in non-invasive tissues[(14.80±1.03)/HPF vs.(3.50±0.53)/HPF,P<0.01].The density of LYVE-1 protein positive lymphatic vessels in invasive tissues was higheRthan that in non-invasive tissues[(25.90±4.43)/HPF vs.(3.60±0.52)/HPF,P<0.01].Conclusion:The expression of HIF-1αprotein is not related to the invasion of lung adenocarcinoma and the density of functional blood vessels and lymphatic vessels,but the density of functional vessels and lymphatic vessels is closely related to the invasion and development of lung adenocarcinoma.
