摘 要:目的探讨半乳糖凝集素3(Gal-3)对肺癌细胞生物学行为及免疫相关因子的影响,并基于Notch信号通路分析其可能的作用机制。方法将A549细胞分为4组:对照组、Gal-3组、LY450139组、Gal-3+LY450139组。对照组转染阴性对照质粒,Gal-3组转染Gal-3过表达质粒,LY450139组给予终浓度为20 nmol/L的Notch信号通路抑制剂LY450139处理,Gal-3+LY450139组给予转染Gal-3过表达质粒和终浓度为20 nmol/L的LY450139处理。采用Western blot检测4组A549细胞Gal-3和Notch受体1(NOTCH1)蛋白表达水平。采用CCK-8法检测4组A549细胞增殖活力。采用流式细胞术检测4组A549细胞凋亡率。采用Transwell法检测4组A549细胞侵袭能力。采用实时荧光定量PCR法检测白细胞介素6(IL-6)和转化生长因子β1(TGF-β1)的mRNA表达水平。结果Gal-3组细胞的Gal-3和NOTCH1蛋白表达水平、增殖活力、侵袭能力、IL-6和TGF-β1 mRNA表达水平高于对照组,而凋亡率低于对照组(均P<0.05);LY450139组细胞的NOTCH1蛋白表达水平、增殖活力、侵袭能力、IL-6和TGF-β1 mRNA水平低于对照组,而凋亡率高于对照组(均P<0.05);Gal-3+LY450139组细胞的NOTCH1蛋白水平、增殖活力、侵袭能力、IL-6和TGF-β1 mRNA水平低于Gal-3组,但高于LY450139组,而凋亡率高于Gal-3组,但低于LY450139组(均P<0.05)。结论Gal-3可通过调控Notch信号通路来增强肺癌细胞的增殖和侵袭能力,并促进免疫相关因子IL-6和TGF-β1的表达。Objective To explore the effect of galectin 3(Gal-3)on biological behavior and immune-related factors of lung cancer cells,and to analyze its possible mechanism based on Notch signaling pathway.Methods A549 cells were divided into 4 groups as follows:control group,Gal-3 group,LY450139 group,or Gal-3+LY450139 group.The control group was transfected with negative control plasmid,the Gal-3 group was transfected with Gal-3 over-expressed plasmid,the LY450139 group was treated using Notch signaling pathway inhibitor LY450139 with final concentration of 20 nmol/L,and the Gal-3+LY450139 group was transfected with Gal-3 over-expressed plasmid and treated using LY450139 with final concentration of 20 nmol/L.The Western blot was used to detect Gal-3 and Notch receptor 1(NOTCH1)protein expressions of A549 cells in 4 groups.The proliferation activity of A549 cells in 4 groups was detected by the CCK-8 method.The apoptosis rate of A549 cells in 4 groups was detected by the flow cytometry.The Transwell method was used to detect the invasion ability of A549 cells in 4 groups.The mRNA expressions of interleukin 6(IL-6)and transforming growth factorβ1(TGF-β1)were detected by the real-time fluorescent quantitative PCR.Results The protein expressions of Gal-3 and NOTCH1,proliferation activities,invasion abilities,and mRNA expressions of IL-6 and TGF-β1 in the cells of the Gal-3 group were higher than those of the control group,while the apoptosis rate was lower than that of the control group(all P<0.05).The NOTCH1 protein expression,proliferation activity,invasion ability,and mRNA expressions of IL-6 and TGF-β1 in the cells of the LY450139 group were lower than those of the control group,while the apoptosis rate was higher than that of the control group(all P<0.05).The NOTCH1 protein expression,proliferation activity,invasion ability,and mRNA expressions of IL-6 and TGF-β1 in the cells of the Gal-3+LY450139 group were lower than those of the Gal-3 group,but higher than those of the LY450139 group,while the apoptosis rate was h
