摘 要:目的:通过生物信息学方法研究胆固醇酰基转移酶-1(acyl-coenzyme A:cholesterolacyltransferase-1,ACAT1)基因在泛癌中的表达情况及临床意义。方法:从TCGA、GTEX及TIMER数据库中获取数据,利用K-M生存分析及Sanger Box进行分析。结果:与正常组织相比,ACAT1基因在泛癌中广泛低表达。ACAT1基因低表达对膀胱尿路上皮癌(bladder urothelial carcinoma,BLCA)、急性髓系白血病(acute myeloid leukemia,LAML)、嗜铬细胞瘤和副神经节瘤(pheochromocytoma and paraganglioma,PCPG)、结肠腺癌(colon adenocarcinoma,COAD)及头颈鳞状细胞癌(head and neck squamous cell carcinoma,HNSC)患者预后较好。在BLCA中,ACAT1基因表达水平与免疫浸润显著正相关,而肾透明细胞癌(kidney renal clear cell carcinoma,KIRC)则相反。ACAT1基因表达水平与泛癌患者免疫治疗的预测标志物(肿瘤新抗原、肿瘤突变负荷、微卫星不稳定性、错配修复缺陷和DNA甲基转移酶)显著相关。结论:ACAT1基因可作为泛癌患者预后及免疫治疗效果的预测标志物,并与免疫浸润有关。Objective:To study the expression and clinical significance of Acyl-coenzyme A:cholesterolacyltransferase-1(ACAT1)gene in pan-cancer through bioinformatics methods.Methods:Data were obtained from TCGA,GTEX and TIMER databases and analyzed using K-M survival analysis and Sanger Box.Results:The ACAT1 gene was widely underexpressed in pan-cancer compared with normal tissues.Low expression of ACAT1 gene was good for bladder urothelial carcinoma(BLCA),acute myeloid leukemia(LAML),pheochromocytoma and paraganglioma(PCPG),colon adenocarcinoma(COAD)and head and neck squamous cell carcinoma(HNSC)patients’prognosis.In BLCA,ACAT1 gene expression level was significantly positively correlated with immune infiltration,but negatively correlated with Kidney renal clear cell carcinoma(KIRC).ACAT1 gene expression levels were significantly associated with predictive markers of immunotherapy(tumor neoantigens,tumor mutational burden,microsatellite instability,mismatch repair deficiency,and DNA methyltransferases)in pan-cancer patients.Conclusion:ACAT1 gene can be used as a predictive marker for the prognosis and immunotherapy effect of pan-cancer patients,and is related to immune infiltration.
