摘  要:为了优化刺糖多孢菌发酵工艺,以刺糖多孢菌YJY-12为试验菌株,以多杀菌素发酵水平为指标,通过正交试验及单因素试验对产多杀菌素的发酵培养基成分及培养条件(种龄、接种量、温度、pH值等)进行优化,确定最佳的培养基组成及培养条件,并对其进行了试验验证。结果显示:最佳的发酵培养基组成为葡萄糖55 g·L^(-1),棉籽蛋白30 g·L^(-1),油酸甲酯40 g·L^(-1),大豆油15 g·L^(-1),酵母粉10 g·L^(-1),蛋白粉15 g·L^(-1),玉米浆干粉5 g·L^(-1),蛋白胨15 g·L^(-1),KH_(2)PO_(4)3 g·L^(-1),(NH_(4))_(2)SO_(4)1 g·L^(-1),CuSO_(4)0.002 g·L^(-1),(NH_(4))_(6)Mo_(7)O_(24)0.002 g·L^(-1),FeSO_(4)0.002 g·L^(-1),THIX-2981 mL·L^(-1),CaCO_(3)5 g·L^(-1)。最适培养条件为pH值7.0,种龄4 d,接种量5%,温度28℃,转速220 r·min^(-1),发酵时间240 h。多杀菌素发酵水平可达2.625 g·L^(-1),较对照组提高了43.99%。综上,研究结果为刺糖多孢菌发酵提供了可靠的发酵培养基配方及发酵工艺,实现了多杀菌素增产目标,为加快多杀菌素的工业化生产提供有益的参考。In order to optimize the fermentation process of Saccharopolyspora spinosa,YJY-12 was used as the test strain,and the spinosad fermentation level was used as the index.The composition of the fermentation medium and the culture conditions(seed age,inoculum amount,temperature,pH,etc)were determined by orthogonal experiments and single factor experiments to determine optimal medium composition and culture conditions.The optimal medium composition and culture conditions were verified by experiment.The results showed that the optimal medium composition was:glucose 55 g·L^(-1),cottonseed protein 30 g·L^(-1),methyl oleate 40 g·L^(-1),soybean oil 15 g·L^(-1),yeast extract 10 g·L^(-1),protein powder 15 g·L^(-1),corn starch powder 5 g·L^(-1),peptone 15 g·L^(-1),KH_(2)PO_(4)3 g·L^(-1),(NH_(4))_(2)SO_(4)1 g·L^(-1),CuSO_(4)0.002 g·L^(-1),(NH_(4))_(6)Mo_(7)O_(24)0.002 g·L^(-1),FeSO_(4)0.002 g·L^(-1),THIX-2981 mL·L^(-1),CaCO_(3)5 g·L^(-1).The optimum culture conditions were pH 7.0,seed age 4 days,inoculation amount 5%,temperature 28℃,rotational speed 220 r·min^(-1) and fermentation time 240 h.The fermentation level of spinosad could reach 2.625 g·L^(-1),which was 43.99%higher than that of the control group.In conlusion,this research provided a reliable fermentation medium formula and fermentation process for the fermentation of Saccharopolyspora spinosa,realized the goal of increasing the production of spinosad,and provided a useful reference for accelerating the industrial production of spinosad.
