摘 要:目的:探讨Siglec⁃9在泛癌中的表达、预后价值、免疫功能及肿瘤进展中扮演的角色。方法:本研究从多个数据库收集转录组、预后及免疫相关信息,可视化分析Siglec⁃9与预后、肿瘤突变负荷(TMB)、微卫星不稳定性(MSI)及肿瘤微环境(TME)的相关性,并进行免疫基因共表达和基因富集分析。结果:Siglec⁃9在大多数肿瘤和癌旁正常组织间有显著的差异表达,并与多种癌症的预后相关。此外,Siglec⁃9还与多种肿瘤的TMB、MSI、TME评分及肿瘤浸润免疫细胞(TIIC)显著相关。基因共表达分析表明Siglec⁃9与多种常见的免疫基因相关,进一步的基因富集分析揭示了Siglec⁃9参与多种免疫相关通路。结论:Siglec⁃9可影响多种癌症的TMB、MSI、TME和TIIC,可作为潜在的癌症预后指标。Objective:To investigate the role of Siglec⁃9 in pan⁃cancer expression,prognostic value,im⁃mune function,and tumor progression.Methods:We collected transcriptomic,prognostic and im⁃mune⁃related information from multiple databases,visualized and analyzed the correlation between Si⁃glec⁃9 and prognosis,tumor mutation burden(TMB),microsatellite instability(MSI),and tumor mi⁃croenvironment(TME),and performed immune gene co⁃relation and gene enrichment analysis.Results:Siglec⁃9 was significantly differentially expressed between most tumors and normal paracan⁃cerous tissues and correlated with prognosis in a variety of cancers.In addition,Siglec⁃9 was signifi⁃cantly associated with TMB,MSI,TME scores and tumor⁃infiltrating immune cells(TIIC)in a vari⁃ety of tumors.Gene co⁃expression analysis showed that Siglec⁃9 was associated with a variety of com⁃mon immune genes,and further gene enrichment analysis revealed that Siglec⁃9 is involved in a vari⁃ety of immune⁃related pathways.Conclusion:Siglec⁃9 can affect TMB,MSI,TME,and TIIC in a variety of cancers and can be used as a potential cancer prognostic indicator.
