摘  要:目的分析五岭龙胆石油醚提取物(PEG)的化学成分,并初步探讨其抗菌、抗炎作用。方法运用气相色谱-质谱(GC-MS)联用仪分析鉴定PEG的化学成分;打孔法测定其对金黄色葡萄球菌、铜绿假单胞菌、大肠杆菌和白色念珠菌的抑菌活性,微量肉汤稀释法测定最低抑菌浓度(MIC);建立脂多糖诱导RAW264.7巨噬细胞炎症模型,给予地塞米松及PEG,检测细胞上清液中NO的含量。结果从PEG中检测出88种成分,鉴定了其中53种,主要为脂肪酸类、角鲨烯等成分;对金黄色葡萄球菌抑菌作用最强,其次为铜绿假单胞菌、大肠杆菌、白色念珠菌,MIC在7.9~31.3 mg/mL。PEG能显著降低脂多糖诱导RAW264.7巨噬细胞炎症中NO的释放。结论PEG主要含有脂肪酸类、角鲨烯等成分,具有较强的抑菌、抗炎作用,其抗菌和抗炎物质基础可能与亚麻酸、亚油酸有关。Objective To study the chemical composition of petroleum ether extract from Gentiana davidii Franch.(PEG), and its antimicrobial and anti-inflammatory effects were also preliminarily discussed.Methods The main chemical composition of PEG was analyzed and identified by gas chromatography-mass spectrometry(GC-MS). The antimicrobial activity of PEG against Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Candida albicans was determined by drilling method. Minimum Inhibitory Concentration(MIC) was determined by micro broth dilution method. RAW264.7 macrophages were stimulated by lipopolysaccharide(LPS) as an inflammation model in vitro, NO content was detected in the supernatant of RAW264.7 cells after treated by PEG and dexamethasone. Results 88 constituents were detected from PEG, and 53 of which were identified. These chemicals are mainly composed of fatty acids. PEG had certain antibacterial activity, and showed strong antibacterial effect against Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Candida albicans. The MIC range was(7.9~31.3) mg/mL. PEG significantly inhibited the content of NO in RAW264.7 cells induced by LPS. Conclusion The PEG contains lots of active component, which have good antibacterial and anti-inflammatory effects that the material basis may be related to linolenic acid, linoleic acid.
