摘 要:目的探讨大麻受体结合蛋白1(cnrip1)调控非洲爪蟾视网膜上皮发育的分子机制。方法利用非洲爪蟾早期胚胎,通过基因敲减观察cnrip1基因表达的下降对视网膜色素上皮发育的影响。同时,采用qRT⁃PCR技术对视网膜色素上皮标记基因otx2、pax6和mitf基因表达情况进行检测以分析cnrip1基因敲减对上述基因表达的影响,随后在这3个标记基因中选择表达量变化最明显的基因进行基因过表达,并与cnrip1过表达所产生的表现型进行对比,以确定cnrip1过表达对视网膜色素上皮发育的影响。结果cnrip1基因敲减导致非洲爪蟾胚胎的视网膜色素上皮的发育受到抑制:使得非洲爪蟾胚胎视网膜上皮的发育发生异常,明显小于对照组otx2、pax6和mitf表达量明显发生下降,与对照组相比,otx2下降至原来的60%,pax6下降至原来的58%,mitf下降至原来71%,即pax6下降得最为显著(P<0.05)。cnrip1基因过表达使视网膜色素上皮向脑部延伸。结论cnrip1对视网膜色素上皮的发育具有调控作用,该作用与otx2、pax6和mitf的表达发生变化有关。Objective To investigate whether and how the cannabinoid receptor interacting protein cnrip1 affects the development of retinal pigment epithelium in Xenopus laevis embryos. Methods By using the early Xenopus laevis embryos, the effect of cnrip1 gene knockdown on the development of retinal pigment epithelium was detected. To determine whether cnrip1 gene knockdown led to the change of marker gene expressions including otx2, pax6 and mitf in the developing retinal pigment epithelium, qRT-PCR was performed. Then the gene whose expression changed most due to cnrip1 gene knockdown was overexpressed and the phenotypes caused was used to compare the phenotypes caused by cnrip1 overexpression. Results Decrease in the size of retinal pigment epithelium was caused by cnrip1 knockdown. The development of the retinal epithelium in Xenopus embryos is abnormal, which is significantly smaller than that of the control, and the expressions of otx2, pax6 and mitf are significantly decreased, compared with the control group. otx2 decreased to 60% of the original, pax6 decreased to 58% of the original, and mitf decreased to 71% of the original, that is, pax6 decreased most significantly(P<0.05). Extension of retinal pigment epithelium to the brain was observed in cnrip1 overexpression experiment. The change of cnrip1 resulted in the changes of otx2, pax6 and mitf expressions. Conclusion The cnrip1 regulates the development of retinal pigment epithelium in Xenopus laevis.
