摘  要:目的:观察牙周炎大鼠肝组织中自噬关键分子酵母Atg6同系物(Beclin1)、微管相关蛋白1轻链3(LC3)及选择性自噬接头蛋白1(SQSTM1/p62)的表达情况,探讨Beclin1介导的自噬在牙周炎相关肝损伤中的作用。方法:随机选取12只雄性Wistar大鼠(8周龄)分为2组,即牙周炎组和对照组(n=6)。牙周炎组采用结扎法建立牙周炎模型,对照组不做处理。8周后检查并记录各组大鼠上颌第一磨牙牙周临床指标,包括龈沟出血指数(SBI)、牙周探诊深度(PD)和牙齿松动度(TM);检测血清中肝功能指标,包括谷丙转氨酶(ALT)、谷草转氨酶(AST)、高密度脂蛋白(HDL)、总胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白(LDL);采用苏木精⁃伊红(HE)染色和油红O染色检测肝组织、牙周组织病理情况及肝组织脂肪性变;实时荧光定量PCR和Western blot检测肝组织中Beclin1、LC3及p62的基因和蛋白表达水平。结果:较对照组,牙周炎组的牙周临床指标(SBI、PD、TM)及HE染色均显示有明显炎症反应;肝功能指标ALT、AST、TC、TG、LDL升高,HDL降低;肝组织HE染色和油红O染色结果显示肝组织内可见空泡样脂肪性变;Beclin1和LC3的mRNA及蛋白表达均降低,p62的mRNA及蛋白表达升高。结论:Beclin1介导的自噬功能障碍可能在牙周炎诱发的大鼠肝损伤中发挥重要的作用。Objective:To observe the expression of Beclin1,microtubule⁃associated protein 1 light chain 3(LC3)and sequesto⁃some 1(SQSTM1)in periodontitis rat liver tissue,and to investigate the role of Beclin1⁃mediated autophagy in periodontitis related liv⁃er injury.Methods:Twelve eight⁃week⁃old male Wistar rats were randomly selected and divided into two groups:the control group and the periodontitis group(n=6).In the periodontitis group,the periodontitis model was established by ligating orthodontic wire,while in the control group,no treatment was given.After 8 weeks,the periodontal clinical indexes of maxillary first molars were examined and recorded,including sulcus bleeding index(SBI),probing depth(PD)and tooth mobility(TM).Also detected were serum liver func⁃tion indicators such as alanine transaminase(ALT),aspartate transaminase(AST),high⁃density lipoprotein(HDL),total cholesterol(TC),total triglycerides(TG),and low density lipoprotein(LDL).HE staining and oil red O staining were used to detect periodontal histopathology,liver histopathology and fatty changes.qRT⁃PCR was used for gene determination of Beclin1,LC3 and p62 in liver tis⁃sue,and Western blot was used for protein determination of Beclin1,LC3 and p62.Results:Compared with the control group,the clinical indicators(SBI,PD,TM)and HE staining of periodontal tissue in the periodontitis group showed obvious inflammatory re⁃sponse.Liver function indicators ALT,AST,TC,TG,LDL increased,but HDL decreased.The results of HE staining and oil red O staining of liver tissue showed vascular steatosis in liver tissue.The mRNA and protein expression levels of Beclin1 and LC3 decreased,while the mRNA and protein expression levels of p62 increased.Conclusions:Beclin1⁃mediated autophagy dysfunction may be part of the mechanisms of periodontitis⁃related liver injury.
