摘  要:为探究不同纯度藻蓝蛋白抗氧化动力学特点,利用2,2′-联氮-双(3-乙基苯并噻唑啉-6-磺酸)(2,2′-azino-bis(3ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基法,测定并建立5种不同纯度藻蓝蛋白清除ABTS阳离子自由基的剂量、时间变化关系,并建立准一级、准二级反应动力学模型,拟合得出反应速率常数,同时分析建立反应速率常数与藻蓝蛋白纯度、色基含量及蛋白质含量的关系.结果发现,5种藻蓝蛋白均能较好地清除ABTS阳离子自由基,其清除率与藻蓝蛋白呈线性量效关系,准二级反应动力学能更好地拟合藻蓝蛋白对ABTS阳离子自由基清除率,且藻蓝蛋白清除ABTS阳离子自由基反应的动力学速率常数与藻蓝蛋白的蛋白质含量纯度成正相关,与藻蓝蛋白的色基相关性不显著或呈负相关,表明藻蓝蛋白对ABTS阳离子自由基起主要清除作用的是藻蓝蛋白中蛋白质部分.In order to explore the antioxidant kinetics of phycocyanin with different purities,we determined and established the relationship between the dose and time of five different purities of phycocyanin scavenging ABTS^(+)·with the method of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)cationic free radical.The pseudo-first-order and pseudo-second-order reaction kinetic models were established,and the reaction rate constant was obtained by fitting.At the same time,the relationship between the reaction rate constant and the phycocyanin purity,chromophore and protein content was analyzed and established.The results showed that the five phycocyanins could scavenge ABTS^(+)·better,and the scavenging rate had a linear dose-effect relationship with phycocyanin,and the pseudo-second-order reaction kinetics could better fit the scavenging rate of phycocyanin to ABTS^(+)·,and the kinetic rate constant of phycocyanin scavenging ABTS^(+)·reaction is positively correlated with the protein fraction and purity of phycocyanin,but has no significant or negative correlation with the chromophore of phycocyanin.It showed that the main scavenging effect of phycocyanin ABTS^(+)·was the protein part of phycocyanin.
