摘  要:作为一种新兴的功能性保健食品,维生素K_(2)受到人们越来越多的关注。传统的诱变育种技术是国内外提高菌种产量和性能的重要手段。以本实验室一株产维生素K_(2)(MK-7)的纳豆芽孢杆菌KVK-09为出发菌株,通过对紫外和氯化锂诱变致死率和正突变率的考察,确定紫外-氯化锂复合诱变的最佳诱变条件为紫外照射30s,氯化锂浓度为0.4%。紫外-氯化锂复合诱变后经过摇瓶初筛和复筛,采用高效液相色谱(HPLC)进行检测并筛选突变株。筛选得到一株维生素K_(2)(MK-7)高产菌株KVK-2694,摇瓶发酵水平达到78μg/mL,是出发菌株(43μg/mL)的1.81倍。采用紫外-氯化锂复合诱变,提高了诱变菌株的正突变率,可以有效提高维生素K_(2)(MK-7)的产量,突变菌株KVK-2694遗传稳定性良好,为工业化生产和应用奠定了良好基础。As a new functional health food,vitamin K,has attracted more and more attention.The technology of traditional mutation breeding is an important means to improve the yield and performance of strains at home and abroad.A strain of Bacillus natto KVK-09 producing vitamin K_(2)(MK-7)in our lab was mutated by UV and LiCl mutation.The optimal conditions of UV-LiCl combined mutation were determined in term of lethality rate and positive mutation rate.The exposure time of UV was determined 30s and the dose of LiCl was determined 0.4%.Primary and secondary assays were made.Content was determined by HPLC method.The mutant strain were isolated.A mutant strain named KVK-2694 was obtained and the yield of vitamin K_(2)(MK-7)reached up to 78μg/mL,which was 1.81 times that of the original strain(43μg/mL).UV-LiCl combined mutation was used to improve the positive mutation rates,which can finally confirm the effectively increase with the yield of vitamin K_(2)(MK-7).The mutant strain KVK-2694 has good genetic stability,which layed a good foundation for the industrial production and application of vitamin K_(2)(MK-7).
