摘 要:目的探索建立茵栀黄注射液过敏反应的BN大鼠动物模型和RBL-2H3细胞模型。方法进行主动全身过敏试验(active systemic anaphylaxis,ASA)试验,比较BN大鼠和Hartley豚鼠的过敏反应症状和反应分级,检测血清IgE和组胺水平;检测茵栀黄注射液作用后RBL-2H3细胞上清组胺释放量,并监测细胞内Ca^(2+)荧光强度。结果过敏反应分级比较,两次攻击后BN大鼠高剂量组较阴性组均有显著性差异(P<0.05),Hartley豚鼠各剂量组较阴性组均无显著性差异;BN大鼠各剂量组组胺水平较阴性组均具显著性差异(P分别<0.01、0.05),豚鼠仅高剂量组较阴性组有显著性差异(P<0.05);RBL-2H3细胞上清中,各剂量组组胺释放量与加药浓度正相关,细胞Ca^(2+)荧光强度的对数与加药浓度的对数正相关。结论BN大鼠评价茵栀黄注射液过敏反应较豚鼠更敏感,血清组胺可作为评价指标;RBL-2H3细胞可用于过敏反应检查的体外模型,细胞上清组胺释放量与细胞Ca^(2+)荧光强度可作为评价指标。Aim To explore the establishment of BN rat animal model and RBL-2H3 cell model of allergic reaction to Yinzhihuang injection.Methods ASA test was performed to compare the symptoms and grades of allergic reactions in BN rats and Hartley guinea pigs,and serum IgE and histamine levels were detected.The amount of histamine released from RBL-2H3 cell supernatant was measured after Yinzhihuang injection affected,and the intracellular Ca^(2+) fluorescence intensity was monitored.Results The grades of allergic reactions were compared.After two attacks,high dose group of BN rats had significant differences compared with negative group(P<0.05),and each dose group of hartley guinea pigs had no significant differences compared with negative group.Each dose groups of BN rats had significant differences in serum histamine levels compared with negative group(P<0.01,0.05 respectively),and only the high dose groups of guinea pigs had significant difference compared with negative group(P<0.05).In the supernatant of RBL-2H3 cells,the amount of histamine released in each dose group was positively correlated with the dosing concentration,and the logarithm of the Ca^(2+) fluorescence intensity of RBL-2H3 cells was positively correlated with the logarithm of the dosing concentration.Conclusions BN rats in evaluation of Yin Zhihuang injection were more sensitive than guinea pigs.Serum histamine levels could be used as indicators of allergic reactions.RBL-2H3 cells could be used as in vitro cell models for allergic reaction tests.The amount of histamine released from the cell supernatant and the Ca^(2+) fluorescence intensity of the cells could be used as indicators for evaluating the degree of allergic reactions.
